藤茶不同提取部位对HepG2细胞胰岛素抵抗改善作用的研究 被引量:2

摘要 目的:研究藤茶不同提取部位改善胰岛素抵抗的活性。方法:采用四甲基偶氮唑盐(MTT)法检测藤茶不同提取部位对Hep G2细胞增殖的影响;用葡萄糖氧化酶法检测藤茶不同提取部位对Hep G2细胞糖消耗的影响;用高糖(55 mmol·L-1)诱导Hep G2细胞,建立胰岛素抵抗模型,并用葡萄糖氧化酶法检测藤茶不同提取部位对胰岛素抵抗Hep G2细胞糖消耗的影响。结果:在有效浓度范围内,藤茶各提取部位对细胞增殖无明显影响,对Hep G2细胞的糖消耗无显著影响。藤茶总提物、正丁醇部位和水部位对胰岛素抵抗Hep G2细胞糖消耗影响显著,与模型组相比,均可使细胞糖消耗量增加50%以上。结论:藤茶不同提取部位对Hep G2细胞糖消耗无显著影响,除石油醚及乙酸乙酯高浓度组(25和50μg·m L-1)外,其余各部位均能提高胰岛素抵抗的Hep G2细胞的葡萄糖消耗,改善Hep G2细胞的胰岛素抵抗。 This article was aimed to study the effects of different fractions from Ampelopsis grossedentata on improving insulin resistance in HepG2 cells. Proliferation of HepG2 cells was detected by 3-(4, 5- dimethyl-2- thiazolyl)-2, 5-diphenyl-2H-terazolium bromide (MTT) assay. Effect of Ampelopsis grossedentata on glucose consumption of HepG2 cells was investigated. Insulin resistance cell model was induced by high concentration of glucose (55 mmol L-1). The hypoglycemic effect of Ampelopsis grossedentata in HepG2 cells was detected. The results showed that different fractions from Ampelopsis grossedentata had little impact on ceil proliferation within the range of effective concentration. There were no obvious influences on glucose consumption of HepG2 cells. Influences of total extract, n-butyl-alcohol fraction and water fraction were remarkable on glucose consumption of insulin resistance in HepG2 cells. Compared with the model group, all of them could increase the glucose consumption for more than 50%. It was concluded that different fractions from Ampelopsis grossedentata, except petroleum ether fraction and ethyl acetate high concentration groups (25 and 50 μg mL-1), could significantly promote the glucose consumption of insulin resistance in HepG2 cells and improve insulin resistance in HepG2 cells.
