摘要
目的:观察菝葜水提物对三硝基苯磺酸(TNBS)诱发大鼠溃疡性结肠炎的疗效,为寻找一种治疗溃疡性结肠炎(UC)的新药提供实验依据。方法:48只SD雄性大鼠随机分成4组,每组12只,分别为正常对照组、模型组、菝葜组、强的松组。除正常对照组,其余各组采用TNBS灌肠法建立UC模型。连续灌胃给药7天后,处死动物,观察结肠组织形态并进行组织学评分,检测大便隐血,HE染色观察病理变化,免疫印迹检测TNF-α的表达,免疫组化、免疫印迹检测NF-κB的表达。结果:菝葜组能改善结肠组织形态,提高组织学评分;菝葜组HE染色显示单核细胞浸润、炎症细胞渗出明显少于模型组;免疫印迹结果显示菝葜水提物能有效降低TNF-α的表达(P<0.01);免疫组化、免疫印迹显示菝葜水提物可有效降低NF-κB的表达(P<0.01),与强的松组效果相当。结论:菝葜水提物对TNBS诱导的UC大鼠具有治疗作用,机制可能是减少TNF-α的表达,抑制NF-κB的激活,从而起到抗炎作用,菝葜可能成为治疗UC的良药。
This study was aimed to observe the effects of Smilax L. water extraction on TNBS induced ulcerative colitis (UC) rats in order to provide animal experiment basis for a new drug in UC treatment. A total of 48 male SD rats were randomly divided into 4 groups, which were the normal control group, model group, Smilax L. group, and the prednisone group, with 12 rats in each group. Except the normal control group, TNBS enema was applied in the UC rat model establishment. Successive intragastric administration was given for 7 days. And then, the rats were sacrificed. Colon tissue morphology was observed and the histological score was evaluated. The fecal occult blood was detected. HE staining was observed for pathological changes. Western blotting was used to detect TNF-a expression. Immunohistochemistry (IHC) and western blotting were used to detect NF-KB expression. The results showed that Smilax L. improved the colon morphological structure and increased the histological score. HE staining of the Smilax L. group indicated that the mononuclear cell infiltration and inflammatory cell exudation were obviously less than the model group. Western blotting showed that Smilax L. water extraction can effectively reduce the TNF-a expression (P 〈 0.01). IHC and western blotting showed that Smilax L. water extraction can effectively reduce the NF-xB expression. There was significant difference compared to the model group (P 〈 0.01). The effect was similar to the prednisone group. It was concluded that Smilax L. water extraction had treatment effect on TNBS induced UC rats. The possible mechanism was to reduce TNF-a expression, inhibit NF-kB expression, in order to have the anti- inflammatory effect. Smilax L. may be a good medicine for UC treatment.
