长春花转化毛状根诱导及培养条件的优化 被引量:37

摘要 用A4和R10 0 0 2种发根农杆菌菌株分别感染长春花的愈伤组织、真叶叶片、叶柄及根 ,诱导出毛状根 .研究了不同外植体、不同菌株以及外植体预培养时间、与发根农杆菌共培养时间、乙酰丁香酮 (As)等条件对转化频率的影响 ,其结果为 :A4转化频率高于R10 0 0 ,真叶叶片转化频率最高 ,最佳预培养时间为 2~ 3d ;最佳共培养时间2~ 3d ;以 10 0mg/mL的As 同时加入共培养基中转化频率高达 86 2 5 % .不同种类的培养基以及碳源。 Transformed hairy roots have been infected in callus, lamina, petioles and roots of catharanthus roseus by Agrobacterium rhizogenes strains A4 and R1000. Effects of different agrobacterium rhizogenes strains,different plants and pre culture, co culture of explants on the transformation frequency were studied. Strain A4, explant lamina, two days pre culture and two days co culture were found the best transformation conditions. The effects of different media, different carbonic, nitrion media on the growth of transformed hairy roots were also studied. 1/2 MS medium contained 3% sucrose as carbonic and 1g/L lactalbumin hydrolysate as nitrion have been found the best conditions for the growth of transformed hairy root.
