烟碱降解菌SCUEC2菌株的分离及其降解特性 被引量:4

摘要 以烟碱为惟一碳源,从湖北省襄阳市烟草种植土壤中分离得到1株烟碱降解菌,为蒙氏假单胞菌(Pseudomonas monteilii)SCUEC2。在30℃下,180 r/min摇床培养,烟碱质量浓度为1.00 g/L时,发酵10 h后烟碱降解率达到88.80%。在30℃下,180 r/min摇床培养48 h,烟碱质量浓度为6.00 g/L时,烟碱降解率为82.59%;烟碱质量浓度为7.00 g/L时,烟碱降解率和菌株生长呈下降趋势,烟碱降解率为18.94%。SCUEC2菌株发酵液经HPLC检测,结果显示,烟碱的保留时间为5.25 min左右,在培养到16 h时,烟碱峰峰面积明显减小,并在保留时间为11.79 min左右处出现一个新色谱峰,表明有中间代谢产物产生。 A nicotine-degradation bacterium,strain SCUEC2,was isolated from the tobacco field soil of Xiangyang City,Hubei Province. Using nicotine as the only carbon source,and it was identified as Pseudomonas monteilii. When the nicotine concentration was 1.00 g/L in the medium,after shaking cultivation for 10 h at 30℃ and 180 r/min,the strain SCUEC2 grew well and degraded 88.80% of nicotine. When the nicotine concentration was 6.00 g/L in the medium, after shaking cultivation for 48 h at 30℃ and 180 r/min,82.59% of nicotine was degraded. When the nicotine concentration was more than 7.00 g/L in the medium, after shaking cultivation for 48 h at 30℃ and 180 r/rain,both the growth and degradation declined,with the degradation less than 18.94%. Analyzing strain SCUEC2 cultured in 1.00 g/L nicotine with HPLC showed that absorption peak existed at 5.25 rain. Peak area decreased gradually and a new peak appeared at 11.79 min after cultivation for 16h,indicating that a intermediate was produced.
