摘要
以珍贵树种土沉香(Aquilaria sinensis)幼茎为材料,研究土沉香启动培养中外植体消毒的最适方法和芽诱导、愈伤组织诱导的最适培养基,植株不同部位的幼茎褐变率比较以及幼茎褐变程度与相关生理指标的关系。结果表明,土沉香启动培养中外植体幼茎的消毒方法以75%的乙醇浸泡30 s、0.1%的升汞浸泡5 min为最佳;芽诱导的培养基以1/2 MS+0.01 mg/L NAA+0.2 mg/L 6-BA为最佳,诱导率为60%;愈伤组织诱导的培养基以MS+0.05 mg/L 2,4-D+2.0 mg/L 6-BA为最佳,诱导率为80%;上部幼茎和下部幼茎相比较,上部幼茎的褐化程度较低。幼茎褐变程度与相关生理指标的关系为:总酚含量越低,PPO活性越低,POD活性越高,褐变越严重。总酚含量下降是导致幼茎褐变的最主要原因。
In this experiment, young stems of the rare species which is Aquilaria sinensis were taken as the research objects, the optimal method of explants disinfection, the optimal medium of bud induction and callus induction, the comparison of browning rate of young stems in different parts of the plant and the relationship between the browning level of young stems and its physiological traits were being research. The results revealed that in startup culture of Aquilaria sinensis, The optimal method of young stems disinfection was with 75% alcohol soaked 30s, 0.1% mercuric chloride soaked 5min; The optimal medium of bud induction was 1/2MS + 0.01 mg/L NAA+0.2 mg/L 6-BA, the induction rate in which was 60%; The optimal medium of callus induction was MS +0.05 mg/L 2 , 4D+2.0 mg/L 6-BA, the induction rate in which was 80%; Compared with young stems in the under parts of the plant, the browning rate of young stems upper was lower. The relationship between the browning level of young stems and its physiological traits was: the lower total phenolic contents/the lower PPO activity/the higher PPO activity, the browning level becomes more serious. The main factor lead to browning of young stems was declining of total phenolic contents.
