摘要 目的:建立高效液相色谱(HPLC)法同时测定癫痫患儿血浆中苯巴比妥(PB)、苯妥荚钠(PH)、卡马西平(CBZ)三种药物浓度。方法:采用HypersllODS2(150mm×4.6mm,5μm)色谱柱,流动相甲醇一水(54:46),流速0.9mlMmin,检测波长240nm,柱温25qc,进样量10μL。结果:PB、PH、CBZ的线性范围分别为1.04~104.24μg/L、0.91—67.99μg/mL、0.37—37.00μg/mL,提取回收率分别为91.50%、89.20%、95.80%。结论:本研究所建立的方法操作简便、结果准确、重现性好,适用于同时测定PB、PH、CBZ三种药物的血药浓度。 Objective : To establish a HPLC method for simultaneous determination of phenobarbital ( PB ), phenytoin ( PH ) and carbamazepine (CBZ) concentrations in human serum of epileptic children. Methods: The separation was performed on a Hypersil ODS2( 150 mmx4. 6 ram, 5 μm)column. The mobile phase was methanol-water( 54 : 46) at a flow rate of 0. 9 mL/min, the column was at 25 ℃, and detected at 240 nm, the injection volume was 10 μL. Results: The calibration curves were linear within the range of 1.04 - 104. 24 μg/mL for PB, 0. 91 ~ 67.99 μg/mL for PH, 0. 37 ~ 37.00 μg/mL for CBZ, and the average recoveries were 91.50%, 89.20%, 95. 80% respectively. Conclusion: This method is simple, accurate, reproducible and suitable for the determination of PB, PH, CBZ blood concentrations of three drugs.
