摘要
配体与受体间的识别及相互作用是分子发挥其功能的重要途径,具有重要的生物学意义。但是,寻找某一分子的受体并不容易,往往成为研究的瓶颈所在。目前寻找及验证某一对配体/受体分子及其对应关系的方法有很多,包括免疫共沉淀、噬菌体展示、酵母双杂交、分子模拟、谷胱甘肽巯基转移酶pull-down、串联亲和纯化等。该文对这些方法的基本原理、简要步骤及优缺点进行综述,为相关研究提供依据。
Recognization and interaction between receptor and ligand is an important way for molecules to perform their functions,and has great biological significance.Researches on receptor and ligand have an essential role in biological science.The progress of numerous researches,however,are frequently prevented by the undiscovered receptors of some specified molecular.The methods to find receptor of a molecular include co-immunoprecipitation, phage display, yeast two-hybrid system, molecular docking simulation, glutathione S-transferase pull-down,and tandem affinity purification.Here is to introduce the fundamental theory,proce-dure,merits and demerits of these methods,in order to provide reference for the related research.
