摘要
杂交子代的纯度直接影响到遗传图谱的质量,因而在遗传图谱构建前要对杂交子代进行鉴定与选择。笔者从300对SSR标记中,筛选出11对能明确显示亲本来源的标记,利用这11对标记对380株美洲黑杨杂交子代进行了鉴定。结果表明:有372个个体因同时具有亲本各1条等位基因而被认定为亲子代可用于图谱构建,其余8个个体因产生了非亲本来源或额外的等位基因,这些个体可能源于花粉污染或染色体异常,在作图过程中宜从作图群体中排除。
Since the purity of hybrids affects the quality of the genetic map, identification and selection of hybrids should be done before the genetic map construction. In this study, we screened 11 SSR pairs primer that could be used to track the kinship of the hybrids from a total 300 pairs primer. Subsequently, these primers were used to test 380 offsprings from a hybridization of Populus deltoides. Among which, 372 individuals were revealed to own complementary alleles for each parent, and they were identified as true hybrids for genetic map construction. Whereas the remaining eight individu?als, they were observed to own non?parental alleles or extra parental alleles, suggesting these progeny were either resul?ted from pollen contamination or chromosomal abnormalities. Thus they were excluded from map construction.
