HPLC法测定软骨素复合胶囊中硫酸软骨素的含量

Determination of the Content of Chondroitin Sulfate in Glucosamine Chondroitin Complex Capsules by High Performance Liquid Chromatography

为建立一种简便准确的测定硫酸软骨素质量分数的高效液相色谱方法，本文采用色谱柱为Kromasil C18柱（4．6×250mm，5μm），流动相为乙腈-10mmol／L戊烷磺酸钠水溶液（10：90），检测波长为192nm，流速为0．8mL／min，进样量为10μl的色谱条件进行实验。实验结果表明，当硫酸软骨素质量浓度在0．1～1．0mg／mL范围内时，其线性关系良好（r=0．9994），平均回收率为99．24％，相对标准偏差为0．85％。该检测方法操作简单，准确度与精密度较高，适用于对硫酸软骨素质量分数的测定，具有一定的推广和应用价值。

To establish a simple and accurate method of high performance liquid chromatography for the de- termination of chondroitin sulfate in glucosamine chondroitin complex capsules, a Kromasil C18 column （4.6×250 mm, 5 μm） was used as a chromatographic column. And the mobile phase was acetonitrile-10 mmol/L sodium pentanesulfonate solution （10 ： 90） at a flow rate of 0.8 mL/min. And the detection wavelength was 192 nm. The sample volume was 10 μL. Experimental results show that the calibration curve was linear in the concentration range of 0.1 ~ 1.0 mg/mL. The average recovery was 99.24% and the relative standard deviation was 0.85 %. This method with good accuracy and high precision is simple and convenient, which is suitable for determination of the mass fraction of chondroitin sulfate. And this determination method has certain value of popularization and application.
