摘要
目的研究高渗盐水(hypertonic saline,HTS)预处理对大鼠缺血再灌注心肌的保护作用及相关机制。方法健康雄性SD大鼠52只,随机分为假手术组(SO组10只)、HTS+SO组(A组10只)、缺血再灌注组(B组10只)、HTS+缺血再灌注组(C组10只)、锌原卟啉IX(ZnPPIX)+HTS预处理组(D组12只)。建立大鼠心肌缺血再灌注模型。检测各组大鼠心肌梗死范围、肌酸激酶(CK)、乳酸脱氢酶(LDH)、TNF-α、白细胞介素6(IL-6)、超氧化物歧化酶(SOD)、丙二醛等的变化。结果与B组比较,C组大鼠心肌梗死体积明显缩小,LDH、CK、TNF-α、IL-6、丙二醛含量明显降低,SOD和HO-1含量明显升高,差异有统计学意义(P<0.05);与C组比较,D组大鼠心肌梗死面积明显增大,LDH、CK、TNF-α、IL-6、丙二醛含量明显升高,SOD含量明显降低,差异有统计学意义(P<0.05)。结论 HTS预处理对大鼠心肌缺血再灌注损伤具有保护作用,其保护机制可能与其抑制炎性因子的释放和调节氧化应激有关。
Objective To study the role of hypertonic saline(HTS)pretreatment in protecting rats against myocardial I/R injury.Methods Fifty-two healthy male SD rats were randomly divided into sham operation(SO)group(n=10),HTS+SO group(n=10),I/R group(n=10),HTS+I/R group(n=10),and ZnPPIX+HTS pretreatment group(n=12).A rat I/R model was established.The myocardial infaction size and serum levels of CK,LDH,TNF-α,IL-6,MDA and SOD were measured in different groups.Results The myocardial infarction size was significantly smaller,the serum levels of CK,LDH,TNF-α,IL-6,and MDA were significantly lower,but the serum HO-1and SOD levels were significantly higher in hypertonic saline+I/R group than in I/R group(P〈0.05).The myocardial infarction size was significantly larger and the serum CK,LDH,TNF-α,IL-6and MDA were significantly higher,but SOD and HO-1levels were significantly lower in ZnPPIX+HTS pretreatment group than in HTS+I/R group(P〈0.05).Conclusion HTS pretreatment can protect rats against myocardial I/R injury by inhibiting the release of inflammatory cytokines and regulating oxidative stress.
