中药柴贝止痫汤对海人酸致痫大鼠脑组织BCRP及MVP表达的调节作用 被引量:10

Regulating effect of Chaibei Zhixian Decoction on expression of breast cancer resistant protein and major vault protein in brain tissues of rats with epilepsy induced by kainic acid

摘要 目的:观察柴贝止痫汤对海人酸致痫大鼠模型脑组织耐药蛋白乳腺癌耐药蛋白(BCRP)、主穹窿蛋白(MVP)表达的作用。方法:本研究采用侧脑室注射海人酸的方法建立癫痫持续状态后慢性难治性癫痫大鼠模型,分为正常组、假手术组、模型组、卡马西平组、中药组和中西药组共6组,给药3个月后取新鲜脑组织分别进行蛋白质印迹法(Western blot)以及RT-PCR来观察各组大鼠脑内BCRP及MVP表达的情况并比较组间差异。结果:正常组、假手术组大鼠皮层的BCRP的表达无明显差异;与假手术组比较,模型组BCRP表达升高(P<0.01);与模型组比较,中药组及中西药组BCRP的表达降低(P<0.05),而MVP的表达中西药组与模型组相比未见明显统计学差异。结论:柴贝止痫汤可降低海人酸致痫大鼠脑组织BCRP的表达,而对于MVP表达的作用还需要进一步的研究论证。 Objective: To observe the effects of Chaibei Zhixian Decoction on expression of breast cancer resistance protein (BCRP) and major vault protein (MVP) in brain tissues of rats with epilepsy induced by kainic acid. Methods: The epilepsy rat models were established by lateral cerebral ventricle injection with kainic acid, and then the rats were randomly divided into six groups as following: control group, sham-operation group, model group, carbamazepine group, Chinese medicine group, and carbamazepine combined with Chinese medicine group. After three-month treatment, the rats were sacrificed, and the brain tissues were obtained and used to observe the differential expression of BCRP and MVP by using Western blot and RT-PCR methods. Results: The difference in expression of BCRP in cerebral cortex of rats between control group and sham-operation group was not significant. Compared with sham-operation group, the expression of BCRP in cerebral cortex of rats in model group was increased significantly (P〈0.01). The expression of BCRP in cerebral cortex of rats in model group was significantly increased compared with Chinese medicine group and carbamazepine combined with Chinese medicine group (P〈0.05). The difference in expression of MVP of rats among carbamazepine combined with Chinese medicine group and model group was not significant. Conclusion: Chaibei Zhixian Decoction could down-regulate the expression of BCRP and increase the concentration of anti-epilepsy drugs of rats with epilepsy induced by kainic acid, however, further studies were need to demonstrate the effect of Chaibei Zhixian Decoction on MVP expression.
