摘要
目的:研究自拟中药方脊髓康对损伤脊髓神经再生和功能重建的机制。方法:采用改良Allen,s法建立大鼠脊髓损伤(SCI)动物模型,随机分为模型组、强的松组、脊髓康高剂量组、脊髓康中剂量组、脊髓康低剂量组共5组,观测每组分别在3、7、14d时的脊髓损伤区细胞凋亡表达与生长相关蛋白-43(GAP-43)表达。结果:术后3d,各脊髓损伤组均出现大量凋亡神经细胞,模型组与其余各组相比差异显著(P<0.01)。术后7d,脊髓康高剂量组及强的松组疗效优于其余各组,两组差异无统计学意义。术后14d,强的松组与脊髓康中剂量组疗效较优,两组差异无统计学意义。各组在3个时间点GAP-43蛋白表达结果均有统计学差异(P<0.05)。与模型组比较,3d时,假手术组、强的松组及脊髓康中剂量组均有统计学差异(P<0.01);7及14d时,各组与模型组相比均有统计学差异(P<0.01)。结论:脊髓康能通过减少SCI后神经细胞的凋亡,上调损伤脊髓中GAP-43的表达,挽救受损的神经元,改善神经恢复的微环境,有利于损伤神经的轴突再生以及神经功能重建。
Objective: To explore the mechanism of Chinese medicine Jisuikang on nerve regeneration and function reconstruction in rats with spinal cord injury. Methods: The rat models of spinal cord injury were established by modified Allen's method. Rats were randomly divided into model group, prednisone group, high, middle and low dosage of Jisuikang groups. The expression of apoptosis and growth-associated protein-43 (GAP-43) was detectedon the 3rd, 7th and 14th day.Results: Numerous apoptotie cells appeared in rats of each group on the 3rd day after the operation, and the difference among model group and the other groups was significant (P〈0.01). On the 7th day after operation, the curative effect of high dosage of Jisuikang group and prednisone group was superior to that of other groups, and the difference between the two groups was not significant. On the 14th day, the curative effect of middle dosage of Jisuikang group and prednisone group was superior to that of other groups, and the difference between the two groups was not significant. For the expression of GAP-43, there were significant differences among each groupin 3 different time points (P〈0.05). Compared with model group, the differences among sham-operation group, prednisone group and middle dosage of Jisuikang group were significant on the 3rd day (P〈0.05). On the 7th and 14th day, the difference among model group and the other groups was significant (P〈0.05). Conclusion: Jisuikang could save injured neurons, improve the micro environment for neurological recovery through reducing neuronal apoptosis and increasing expression of GAP- 43 after spinal cord injury, which was helpful for axonal regeneration in injured nerve and neural function reconstruction.
