摘要
目的:探讨临床经验方固本防哮饮对哮喘缓解期小鼠白细胞介素-12(IL-12)及免疫球蛋白E(IgE)表达的影响。方法:采用卵蛋白(OVA)致敏和激发联合呼吸道合胞病毒(RSV)滴鼻的方法,建立BALB/c小鼠哮喘缓解期模型,分别灌胃给药,分为正常组,模型组,固本防哮饮中、高、低剂量组及孟鲁司特钠组。ELISA法检测各组小鼠血清中的IgE水平和肺泡灌洗液中IL-12的表达水平。结果:模型组小鼠血清中IgE的平均含量显著高于正常组(P<0.01);肺泡灌洗液中IL-12平均含量显著低于正常组(P<0.01)。孟鲁司特钠组及固本防哮饮各剂量组小鼠IgE的平均含量均低于模型组(P<0.05);孟鲁司特钠组IL-12的平均含量显著高于模型组(P<0.05),固本防哮饮各剂量组IL-12的平均含量高于模型组,但差异无统计学意义。结论:IL-12的分泌不足及IgE的高表达可能与缓解期慢性炎症持续存在有关。而固本防哮饮可通过升高IL-12水平,减少IgE的合成,改善气道慢性炎症,从而减轻或预防哮喘的发作。
Objective: To investigate the effects of Guben Fangxiao Decoction on expression of interleukin-12 (IL-12) and immunoglobulin E (IgE) in mice with asthma remission. Methods: BALB/c mice model with asthma remission was established by using the methods of ovalbumin (OVA) sensitization combined with respiratory syncytial virus (RSV) nasal inhalation, and the mice in control group, model group, montelukast group, and high, middle, low dose of Guben Fangxiao Decoction groups. ELISA was used to test the IgE level in mice serum and IL-12 level in pulmonary alveolar lavage fluid. Results: The average level of IgE of mice in model group was significantly higher than that in control group (P〈0.01). The average level of IL-12 in pulmonary alveolar lavage fluid was lower than that in control group (P〈0.01). The average level of IgE of mice in montelukast group and every-dosage group of Guben Fangxiao Decoction were lower than that in model group (P〈0.01). The average level of IL-12 of mice in montelukast group was higher than that in model group (P〈0.05). The average level of IL-12 of mice in each dosage Decoction group was higher than that of model group, but there was no significant difference. Conclusion: The hyposecretion of IL-12 and the high expression of IgE might be associate with the persistent chronic inflammation of paracmasis. Guben Fangxiao Decoction could improve the chronic inflammation of airway and reduce or prevent the onset of asthma by increasing IL-12 level and reducing compound of IgE.
