摘要
目的研究分析硫酸沙丁胺醇对秀丽线虫的毒性作用。方法以不同浓度的硫酸沙丁胺醇暴露野生型秀丽线虫N2的L1幼虫至年轻成虫(young adult),以K缓冲液作为空白对照,通过检测暴露后秀丽线虫的存活率、运动行为及其后代数目等生理指标,对硫酸沙丁胺醇的毒性进行分析。结果与空白对照组比较,实验浓度范围内硫酸沙丁胺醇对秀丽线虫没有致死作用,从浓度0.01 mg/L开始对秀丽线虫产生毒害作用,表现为身体弯曲、头部摆动频率降低,后代数量减少等;进一步检测F1代线虫的运动行为和生殖情况,发现F1代也表现出相似的症状,说明硫酸沙丁胺醇对秀丽线虫的毒性是可遗传的;以同样的浓度暴露线虫突变体daf-16,也得到相似的结果,而且daf-16比野生型N2反映更灵敏。结论硫酸沙丁胺醇对秀丽线虫N2及daf-16的运动行为和生殖均有毒害作用。
Objective To analyse the toxicity of Salbutamol Sulfate by using a model organism C.elegans. Methods The C.elegans were treated with different dose of Salbutamol Sulfate,and the control group were treated with k-medium. The toxicity was detected using the indicators of suivial,body bends, head thrashes and brood size. Results Compared with control group,the dose could not kil ed the C.elegans,but the indicators of body bends,head thrashes and brood size were decreased from the dose of 0.01 mg/l. Furthermore,we obtained similar results for the F1 generation. So the toxicity of Salbutamol Sulfate to C.elegans was heritable. When the same treatments to mutant Daf-16,we found the similar results,and the mutant was more sensitive to the treatments. Conclusion Salbutamol Sulfate is toxic to the locomotion behavior and reproduction of C.elegans.
