摘要
目的检测24-脱氢胆固醇还原酶(3β-hydor-xysteroid-△24 reducase,DHCR24)在结直肠癌及癌旁肠黏膜中的表达,探讨其临床意义。方法应用组织芯片技术结合免疫组化法检测第三军医大学3所附属医院结直肠癌术后125例组织标本以及相应63例癌旁肠黏膜中DHCR24的表达。结果DHCR24在125例结直肠癌组织中阳性表达率显著高于癌旁肠黏膜(67.2%vs 39.7%,P<0.01),并与肿瘤的分化程度、临床分期、淋巴结转移等有关(P<0.05),结直肠癌患者中DHCR24高表达组比低表达组预后差(P<0.01),临床分期高的结直肠癌患者中,DHCR24的高表达提示了临床预后差(P<0.01),但在临床分期低的患者中不存在这种差异(P>0.05)。结论 DHCR24在结直肠癌组织中高表达,且结直肠癌患者中DHCR24高表达组比低表达组预后差,提示其在结直肠癌的发生、发展中可能发挥着重要作用。
Objective To determine the expression level of 313-hydor-xysteroid-△24 reducase (DHCR24) in colorectal cancer and adjacent mucosa tissues, and explore its clinical significance. Methods Tissue microarray and immunohistochemical staining was applied for the detection of DHCR24 expression in 125 cases of colorectal cancer and 63 adjacent mucosa tissues through surgical treatment. The results were analyzed with the clinical data. Results The positive expression rate of DHCR24 was significantly higher in the colorectal cancer tissue than in the adjacent mucosa tissue ( 67.2% vs 39.7% , P 〈 0.01 ). DHCR24 expression was significantly correlated with tumor differentiation degree, clinical stage and lymph node metastasis (P 〈0.05). The patients with high expression of DHCR24 had poorer prognosis (P 〈 0.01 ). Among the patients who had advanced-stage colorectal cancer, higher DHCR24 expression suggested a poorer survival ( P 〈 0.01 ). But no such suggestion was seen in the patients at early stage disease ( P 〉 0.05 ). Conclusion DHCR24 is highly expressed in eolorectal cancer tissue, and the patients with high expression predicts poorer prognosis. DHCR24 might play an important role in the malignant progression of colorectal cancer.
