ppGalNAc-T1和-T2在乳腺癌相关药物敏感性、细胞凋亡及侵袭中的作用

Role of ppGalNAc-T1 and ppGalNAc-T2 in chemosensitivity,apoptosis and invasion of breast cancer cells

摘要 目的探讨N-乙酰氨基半乳糖转移酶(ppGalNAc-T)1和-T2对三阴性乳腺癌细胞株MDA-MB-231恶性表型的影响。方法分别将siRNA-ppGalNAc-T1(A组)、siRNA-ppGalNAc-T2(B组)和空载体对照(C组)转染MDA-MB-231细胞。采用荧光定量PCR检测ppGalNAc-T1和-T2mRNA表达,MTT法和Hoechst 33258染色分别检测各组对多西他赛和吉西他滨的药物敏感性及细胞凋亡率的变化,Transwell实验检测各组细胞的侵袭能力。结果MDA-MB-231细胞中ppGalNAc-T1和-T2mRNA表达水平均高于正常乳腺细胞(P<0.05)。与C组相比,A组、B组药物敏感性和细胞凋亡率升高(P<0.05),而细胞侵袭能力受到抑制(P<0.05)。结论 ppGalNAc-T1和-T2可能是潜在预测和治疗三阴性乳腺癌的有效靶点。 Objective To investigate the effect of polypeptide N-acetylgalactosaminyltrans- ferase(ppGalNAc)-T1 and ppGalNAe-T2 on malignant phenotype of triple-negative breast cancer cells. Methods MDA-MB-231 cells were transfected with siRNA-ppGalNAe-T1 (group A), siRNA- ppGalNAe-T2(group B) and empty vector(group C). The mRNA expressions of ppGalNAe-T1 and ppGalNAc-T2 were determined by fluorescence quantitative-PCR. The chemosensitivities and apoptosis of docetaxel and gemcitabine after transfection were detected by MTT and Hoechst 33258 staining, respectively. And cell invasion was measured by Transwell experiment. Results The mRNA expressions of ppGalNAc-T1 and ppGalNAe-T2 in MDA-MB-231 cells were higher than those in normal breast cells (P〈0. 05). Compared with group C, the chemosensitivities and cell apoptosis were increased( P〈0. 05 ), while cell invasion was inhibited in groups of A and B (P〈0. 05). Conclusion ppGalNAe-T1 and ppGalNAe-T2 may be taken as the potential targets for predicting and curin~ triple-negative breast cancer.
