子痫前期患者胎盘组织中TNF-α和MSTN的表达差异及临床分析 被引量:5

摘要 目的探讨轻、重度子痫前期患者和正常产妇胎盘组织中TNF-α和MSTN的表达差异与子痫前期发病程度的关系。方法选择足月健康产妇40例为正常妊娠组、确诊轻度子痫前期产妇40例作为实验A组、重度子痫前期产妇40例为实验B组。免疫组化SP法检测TNF-α和MSTN在胎盘组织中的定位,反转录-半定量PCR(RT-PCR)及蛋白印迹法(Western blot)检测胎盘组织中TNF-α和MSTN mRNA和蛋白的表达差异。结果各组胎盘组织中均有TNF-α和MSTN表达,位置基本相同。TNF-α和MSTN mRNA和蛋白的表达水平在A组和B组明显高于对照组,差异有统计学意义(P<0.01)。B组显著高于A组,差异有统计学意义(P<0.01)。结论子痫前期患者胎盘组织中TNF-α和MSTN的分子生物学的变化,可能是子痫前期病情加重的机制之一。 Objective To detect the relationship of the expression of TNF alpha and MSTN in placenta tis- sue and preeclampsia. Methods 40 cases of full-term healthy maternal (normal group), 40 cases of mild pre- eclampsia women ( experimental group A) and 40 cases of severe preeclampsia women (experimental group B) were recruited in this study. Expression and location of TNF alpha and MSTN in the placenta tissue were determined by irrmaunohistochemical SP method, semi-quantitative PCR (RT- PCR) and protein imprinting methods (Western blot). Results TNF alpha and MSTN expressed in all the three groups and the location in placenta is basically the same. Expression levels of TNF alpha and MSTN mRNA was significantly higher in group A and group B compared to control group (P〈0.01) and they were significantly higher in Group B than in group A (P〈0. 01). Conclusion The changes of the molecular biology of MSTN and TNF-α may be one of the mechanism of preeclampsia.
