摘要 神经生长因子(NGF)是一种多效性神经营养因子,与特异性酪氨酸激酶受体结合,介导一系列生物学效应。除了在神经系统的生长发育过程中起关键作用外,神经生长因子及其功能性受体酪氨酸蛋白激酶A(tyrosine kinase A,Trk A)在肿瘤中也起了重要作用。研究表明,NGF-Trk A与肿瘤发生、增殖、血管生成以及转移相关。以Trk A受体为靶点,应用Trk A抑制剂阻断其信号通路可为肿瘤治疗提供新方法,且目前已有部分药物进入临床试验。本文就NGF-Trk A信号通路在肿瘤中的作用以及其靶向治疗的研究进展进行综述。 Nerve growth factor(NGF), a pleiotropic molecule, can bind to specific tyrosine kinase receptor Trk A and induce a number of biological activities. NGF and Trk A are involved not only in the development of nervous system, but also in the pathogenesis of different types of malignancies. Studies have shown that NGF and its Trk A receptor are closely related to carcinogenesis, proliferation, angiogenesis and metastasis of solid tumors. Thus, inhibitors targeting at Trk A may provide a new approach for cancer treatment, and some of them have already been applied in clinical trials. This review summarizes the involvement of NGF-Trk A in tumors and related progress of molecular targeted therapies.
