麻枳降浊方对便秘型肠易激综合征大鼠肠道间质细胞的影响 被引量:12

Effect of Mazhi Jiangzhuo Fang( 麻枳降浊方) on Intestinal Interstitial Cell of Rats with Constipation Type of Irritable Bowel Syndrome

摘要 目的探讨麻枳降浊方治疗便秘型肠易激综合征(IBS-C)的可能作用机制。方法将60只Wistar大鼠随机分为正常组10只和造模组50只,造模组采用冰水灌胃法建立IBS-C模型,将造模成功的48只大鼠随机分为模型组9只、阳性药组10只、高剂量组10只、中剂量组10只、低剂量组9只,阳性药组给予西沙必利片3.6 mg/(kg·d)灌胃,高、中、低剂量组分别给予麻枳降浊方颗粒剂25.2、12.6、6.3 g/(kg·d)灌胃,模型组给予等量蒸馏水灌胃,正常组正常饲养。14天后,HE染色观察肠组织病理变化,免疫组织化学法检测肠组织Cajal间质细胞(ICC)数量和形态的变化,并采用Image-Pro Plus 6.0图像分析系统分析c-kit基因阳性表达面积和积分光密度(IOD)。结果模型组大鼠肠道ICC细胞突起变短,带状、网状结构不明显,各给药组大鼠肠组织ICC数量、形态、分布及网状连接与正常组相似。模型组大鼠c-kit的阳性表达面积及IOD较正常组显著减少(P<0.05)。高、中剂量组和阳性药组c-kit阳性表达面积及IOD较模型组增多(P<0.05),并且高、中剂量组显著高于阳性药组(P<0.05)。结论麻枳降浊方可能通过增加肠组织ICC细胞数量,改善ICC形态结构,增加肠道传导功能,从而改善IBS-C。 Objective To explore the possible mechanism of Mazhi Jiangzhuo Fang in the treatment of irritable bowel syndrome( IBS-C). Methods 60 Wistar rats were randomly divided into normal group( n = 10) and modeling group( n = 50). Rat models of IBS-C were established with ice water gavage method in modeling group. 48 completed modeled rats were randomly divided into model group( n = 9),positive drug control group( n = 10),high dose group( n = 10),middle dose group( n = 10) and low dose group( n = 9). Positive drug control group was given Cisapride tablets( 3. 6 mg /( kg ·d)) by gavage. High,middle and low dose groups were given Mazhi Jiangzhuo Fang granules 25. 2 g /( kg·d),12. 6 g /( kg·d) and 6. 3 g /( kg·d) by gavage,respectively. Model group was given equivalent distilled water by gavage. Normal group was fed normally. After 14 days,the pathological changes of intestinal tissue was observed by hematoxylin and eosin stain( HE stain). Changes in amount and shape of interstitial cell of Cajal( ICC) were detected. The positively expressed proportion of c-kit gene and integral optical density( IOD) was analyzed with image analysis system of Image-Pro Plus 6. 0. Results The protuberances of ICC in model group rats were shorten. Banding and reticulation structure were not obvious. The amount,shape,distribution and reticulated connection of rats’ ICC in medication administration groups were similar to those in normal group. The positively expressed proportion of c-kit gene and IOD in model group were less than those in normal group( P 〈0. 05). The positively expressed proportion of c-kit gene and IOD in high and middle dose groups and positive drug control group were more than those in model group( P 〈 0. 05),especially in high and middle dose groups( P 〈0. 05). Conclusion Mazhi Jiangzhuo Fang may improve IBS-C by increasing the amount of ICC,improving the shape and structure of ICC,and strengthening the rhythmic contraction of intestinal tract.
