摘要
目的探讨通络醒脑泡腾片(TLXNET)对阿尔茨海默病(AD)模型大鼠海马PDH和α-KGDHC表达的影响。方法建立淀粉样蛋白片段(Aβ25-35)海马注射和双侧颈总动脉永久结扎两种大鼠AD模型。用Morris水迷宫法检测大鼠的学习记忆能力;免疫组化法定量分析大鼠海马丙酮酸脱氢酶(PDH)和α-酮戊二酸脱氢酶(α-KGDHC)蛋白的表达。结果与模型组比较,TLXNET能显著延长两种AD模型大鼠训练第5天的逃避潜伏期,改善空间探索行为;能显著提高大鼠海马PDH和α-KGDHC的表达,提高平均光密度值(IA);但在Aβ25-35海马注射模型中,TLXNET低剂量组的与模型组的比较无显著差异。结论 TLXNET可改善AD模型大鼠的学习记忆能力,其机制可能与促进PDH和α-KGDHC的表达,增加脑内糖代谢的水平,提高脑能量的供应有关。
OBJECTIVE To study the effects of Tongluo Xingnao effervescent tablet( TLXNET) on the expression of hippocampus PDH and α-KGDHC in Alzheimer's disease( AD) rats. METHODS Two kinds of AD rat models were established by Aβ25-35 hippocampus injection and permanent bilateral common carotid arteries occlusion. Learning and memory ability were evaluated by the Morris water maze,and the hippocampal PDH and α-KGDHC expression with immunohistochemistry analysis were also examined.RESULTS TLXNET increased escape latency on the fifth day and improved space exploration ability,meanwhile it also increased the expression of hippocampal PDH and α-KGDHC and average optical density in the two models. All the mentioned above had statistical significance compared with model control group except for low-dose group in the AD rats with Aβ25-35 hippocampus injection.CONCLUSION TLXNET can improve learning and memory of two kinds of AD model rats which suggests that its potential role was related to increasing the level of brain glucose metabolism and enhancing brain energy supply through promoting PDH and α-KGDHC expression.
