不同针刺刺激量对寒凝证类痛经模型大鼠中枢阿片受体影响的研究 被引量:14

Effect of Different Acupuncture Stimulus on Opioid Receptors of Midbrain and Spinal Cord Tissue of Dysmenorrhea Rats with Cold Syndrome

摘要 目的:观察针刺动物不同"得气"状态下,对中枢神经系统内阿片受体mRNA表达的影响,探讨动物得气的不同调节机制。方法:动情间期SD雌性大鼠44只,随机分为盐水对照组、寒凝证类痛经模型组(模型组)、刺激量A组(期望"得气"组)与刺激量B组(期望"不得气"组),除盐水组外其余各组均采用全身冷冻加雌二醇注射方法制备类痛经大鼠模型,刺激量A组诱导动物"得气"状态采用粗针、深刺、行手法;刺激量B组偏于避免"得气"的干预方式即细针、浅刺、不施手法,留针20 min后采用RT-PCR法半定量检测大鼠中脑及脊髓组织内源性阿片肽系统κ、μ受体基因表达的变化。结果:1中脑κ受体mRNA的表达:与盐水组比较模型组表达显著减弱,与模型组比较刺激量A组表达显著增强,与刺激量A组比较刺激量B组表达显著减弱(P<0.05);2中脑内μ受体mRNA的表达:与盐水组、模型组比较,刺激量A组表达显著增强(P<0.05或P<0.01);3脊髓内κ受体、μ受体mRNA的表达:与盐水组比较刺激量A组及刺激量B组κ、μ受体表达均显著增强,P<0.05;与模型组比较刺激量A组κ、μ表达均显著增多,P<0.05。结论:不同"得气"状态下镇痛机制存在差异,刺激量A组能通过增强寒凝证类痛经模型大鼠中脑及脊髓内阿片受体表达起到镇痛效应,且整体较刺激量B组增强更为显著。 Objective:To investigate the effect of different acupuncture methods at SP6 on opioid receptors of brain and spinal cord of primary dysmenorrheal model rats with cold syndrome .Methods:44 female SD rats with 3 months aged were randomized into 4 groups,including a saline control group,a cold stagnation of primary dys-menorrheal model control group( a model control group) ,an A stimuli group( Deqi group) and a B stimuli group ( no Deqi group) .The model groups were with the whole body freezing method combined with estradiol benzoate injection mold,and the saline control group rats were injected 0.9%Nacl.A stimuli group used needling bilat-eral SP6 acupoint for reducing Deqi.B stimuli group used needling bilateral SP6 acupoint,puncturing vertically&amp;nbsp;with one 0.18 &#215;13 mm stainless,sterilized and disposable to a depth of 1-2 mm,and maintaining the needling in place for 20 minutes.Observe the κand μopioid receptor mRNA expression in midbrain and spinal cord of rats in each group.Results:In Midbrain:Compared with the saline group,in the model group the κopioid re-ceptor mRNA expression was significantly decreased( P〈0.05) .Compared with the model group and B stimuli group, theκopioid receptor mRNA expression in A stimuli group was increased significantly( P〈0.05) .Com-pared with the model group,the μopioid receptor mRNA expression in A stimuli group was increased signifi-cantly(P〈0.01).In Spinal:Compared with the saline group, the opioid receptor in the model group was in-creased;compared with the model group,theκandμopioid receptor mRNA expressions in A stimuli group were increased significantly( P〈0.05 ) .Conclusion:The analgesic mechanisms of different“Deqi” status in opioid receptors were different.The A stimuli group can significantly increase midbrain and spinal cord of opioid recep-tors of dysmenorrhea rats with cold syndrome.
