摘要
目的:探讨电针曲池穴、神门穴、足三里穴对心肌肥厚模型大鼠Bcl-2及Bax影响。方法:50只Wistar大鼠随机分为模型组、假手术组、曲池+足三里组、神门+足三里组、曲池+神门+足三里组,每组10只,胸主动脉缩窄法建立大鼠心肌肥厚模型,电针组连续治疗14天,通过实时定量PCR及Western blot测定心肌组织中Bcl-2、Bax mRNA及蛋白的表达水平。结果:曲池+足三里组、神门+足三里组、曲池+神门+足三里组Bcl-2及Bax mRNA及蛋白的表达水平在治疗后与模型组相比,有显著性差异(P<0.05)。与模型组对比各电针组血流动力学均有明显改善。结论:同时电针曲池穴+神门穴+足三里穴可明显抑制胸主动脉缩窄法所引起的心肌肥厚,其机制可能是通过改善心脏血流动力学、以及促进Bcl-2的表达同时抑制Bax的表达进而抑制心肌细胞的凋亡,从而产生延缓及抑制心肌肥厚进展的作用。
Objective:To discuss the effect of electroacupuncture at Quchi, Shenmen and Zusanli on Bcl-2 and Bax in rats with cardiac hypertrophy.Methods:Fifty Wistar rats were randomly divided into five groups ( a model group,a sham operation group,a Quchi &amp; Zusanli group,a Shenmen &amp; Zusanli group and a Quchi &amp;Shenmen &amp;Zusanli group) , including ten rats in each group.The rat models with cardiac hypertrophy were prepared and rats in electroacupuncture groups received a continuous wave for the treatment of 2 weeks. Through the method of real time PCR and western blot, the expression levels of Bcl-2 and Bax mRNA and protein in heart tissue were detected.Results:After treatment,the expression levels of Bcl-2 and Bax mRNA and protein in heart tissue were significantly different in Quchi &amp; Zusanli group, Shenmen &amp; Zusanli group and Quchi &amp; Shenmen&amp;Zusanli group compared with model group(P〈0.05).The hemodynamics of elec-troacupuncture groups improved significantly compared with model group.Conclusion: Electroacupuncture at Quchi &amp;Shenmen&amp;Zusanli acupoints can prevent the myocardial hypertrophy.The mechanism may be by im-proving cardiac hemodynamics and promoting the expression of Bcl-2 and inhibiting the expression of Bax to inhibit the apoptosis of myocardial cells,then playing a role in delaying and inhibiting the process of myocardi-al hypertrophy.
