不同剂量地佐辛复合右美托咪定的镇痛及镇静效果的比较 被引量:20

摘要 目的比较不同剂量地佐辛复合右美托咪定应用于小鼠的镇痛及镇静效果。方法选取ICR品系雌性小鼠80只,随机分为8组(n=10),10μg·kg-1右美托咪定组(DEX组)、0.3 mg·kg-1地佐辛组(D1组)、1.0 mg·kg-1地佐辛组(D2组)、3.0 mg·kg-1地佐辛组(D3组)以及10μg·kg-1右美托咪定分别复合0.3,1.0,3.0 mg·kg-1地佐辛(C1组、C2组、C3组)和生理盐水对照组(NS组)。腹腔注射各组药物15 min后,先采用热板实验观察镇痛效果,继而应用自发活动视频分析系统记录小鼠的活动轨迹,根据小鼠用药后1 h的总路程推断其镇静程度。结果 (1)热板实验:与NS组相比,DEX、D1组给药后热痛阈差异无统计学意义(P>0.05),其他5组热痛阈均高于NS组(P<0.05);复合组热痛阈明显高于单用相应剂量地佐辛及DEX组(P<0.05)。(2)旷场试验:与NS组相比,单用地佐辛各组小鼠总路程无明显差异(P>0.05),DEX组明显短于NS组(P<0.05);复合组明显短于NS组(P<0.05),而与DEX组相比差异无统计学意义(P>0.05),与单用相应剂量的地佐辛相比,总路程均明显减少(P<0.05)。结论地佐辛与右美托咪定复合应用镇痛效果增加,而镇静效果不叠加。 Obiective To compare analgesia and sedation effects of different doses of dezocine combined with DEXmedetomidine in mice.Methods 80 female ICR (Institute of Cancer Research)mice were randomly assigned into 8 groups (n=10 each):10 μg· kg-1 DEXmedetomidine group (DEX group),0.3 mg·kg-1 dezocine group (D1 group),1 .0 mg·kg-1 dezocine group (D2 group), 3.0 mg·kg-1 dezocine group (D3 group)and 10 μg·kg-1 DEXmedetomidine plus 0.3,1.0,or 3.0 mg·kg-1 dezocine group (C1 group,C2 group,C3 group)and 0.9 %NaCl group (NS group).The hot plate test was used to observe the analgesia effects 15 minutes after intraperitoneal injection,and then the locomotion trail of mice was recorded through open field test.The degree of sedation was e-valuated based on the total distance covered one hour after administration.Results 1 .Hot plate test:there was no significant difference in pain threshold in DEX group and D1 group (P〉0.05)compared with NS group;the pain threshold in the other five groups was sig-nificantly higher than NS group (P〈0.05 );the analgesic effects of the two drugs combination are significantly better than the single drug group(P〈0.05).2.Open field test:After injection of different doses of dezocine,the total distance had no obvious difference com-pared with the control group (P〉0.05),while it was shortened in DEX and combined groups (P〈0.05).Conclusions Compared with the groups of dezocine or DEXmedetomidine alone,the effects of analgesia was better in the dezocine combined with DEXmedeto-midine group;Dezocine and DEXmedetomidine have synergistic analgesia effects,but not sedation effects.
