7-乙基-10-羟基喜树碱制备及药动学与毒性

摘要 目的:通过对7-乙基-10-羟基喜树碱白蛋白纳米粒进行制备来提高药物在人身体内部的循环时间并将其中所蕴含的毒性进行降低。方法:采用高压均质的方法对SN38的白蛋白纳米粒进行制备,并且,对制剂的粒径、形态学以及其包封率进行探究,在此同时,应用LC-MS/MS对其在老鼠血浆内药物的浓度进行测定,并且通过对老鼠外周的血白细胞的数量进行检查来对其骨髓抑制毒性的情况进行评定。结果:制备的纳米粒大小均匀,形态良好,包封率为88.14%±3.15%,粒径为16.23±5.9nm,PDI为0.137±0.015;大鼠尾静脉iv给药后,SN38-HSA-LN较SN38溶液的tl/2延长,AUC显著增加,且骨髓抑制程度明显降低。结论制备的SN38-HSA-LN包封率较高,粒径大小分布均匀,静脉注射延长了药物血液循环时间,并降低了骨髓抑制毒性。
