摘要
目的检测大鼠角膜碱烧伤后角膜及房水中缺氧诱导因子(hypoxia-induciblefactor,HIF)-1α的表达情况,探讨角膜碱烧伤后早期HIF-1α在其损伤机制中的作用。方法选择健康成年SD大鼠25只,右眼建立大鼠角膜碱烧伤模型为实验组,左眼不作处理为正常对照组,免疫组织化学法检测碱烧伤后1 d、4 d、7 d、10 d、14 d HIF-1α在角膜中的表达情况,ELISA法检测各时间点房水中HIF-1α的表达量。结果正常对照组中,HIF-1α在角膜及房水中均无表达。实验组碱烧伤后,HIF-1α在角膜上皮细胞层和基质层组织中的表达均较对照组增强,且随碱烧伤时间延长而逐渐增强,至14 d时达到峰值。房水中HIF-1α在碱烧伤后1 d的表达量为(0.056±0.013)μg·L-1,随后持续增高,至14 d时达(0.823±0.114)μg·L-1。房水中HIF-1α的表达量与其在角膜中的表达量呈正相关(r=0.87,P<0.01)。结论 HIF-1α参与了角膜损伤的病理机制,其表达变化与角膜新生血管的生长情况相关。
Objective To detect the expression of hypoxia-inducible factor-Ice (HIF-1α) in the cornea and aqueous humor after corneal alkali burn in rat,and investi- gate the role of HIF-1α in the early mechanisms of injury after corneal alkali burns. Methods Twenty-five healthy adult SD rats were selected,the model of corneal alkali burn in right eye was established as the experimental group, the left eyes received no any treatment as the control group,the expression of HIF-lα at 1 day,4 days,7 days,10 days, 14 days after alkali burn in the cornea were detected by immunohistochemical as- say,and the expression level of HIF-1α in aqueous humor were examined by ELISA. Resultu In the control group,HIF-1α had no expression in the cornea and aqueous humor, while after the alkali burns its expression enhanced in the corneal epithelial cells and matrix layer,which peaked at 14 days. HIF-la expression level in the aqueous hu- mor at 1 day after alkali burn was (0. 055 _+0. 013) μg · L-1 ,and then continued to in- crease,at the 14 days was (0. 823 +0. 114) μg · L-1. Meanwhile,HIF-lμ expression level in the aqueous humor was positively correlated with its expression in cornea ( r = 0.87, P〈0. 01). Conelusion HIF-la play a role in the corneal injury, its expressions change is related with the growth of corneal neovascularization.
