XIAP在子痫前期患者胎盘血管内皮细胞中的表达及意义 被引量:4

The expression of Xchromosome linked inhibitor of apoptosis protein in placental villous endothelial cells of preeclampsia

摘要 目的探讨凋亡抑制蛋白XIAP在子痫前期患者胎盘绒毛血管内皮细胞中的表达及绒毛血管密度的改变。方法收集2013年4至12月山西医科大学第二医院产科收治的30例子痫前期患者(研究组)及30例正常晚期妊娠妇女(正常对照组)的胎盘组织及全部临床资料,用免疫组织化学二步法检测XIAP在胎盘绒毛血管内皮细胞中的定位及Image-Pro Plus医学图像分析系统对图片进行处理,并测定光密度值进行半定量分析。同时进行HE染色,计数绒毛血管数。统计学分析应用SPSS 17.0软件进行独立样本t检验。结果 (1)两组临床资料的比较:研究组与正常对照组在年龄上无显著性差异(P>0.05);在收缩压、舒张压、孕龄、新生儿出生体重有显著性差异(P<0.05)。(2)免疫组化检测结果:研究组和正常对照组在胎盘绒毛血管内皮细胞的胞质中均可见XIAP的阳性表达,且在子痫前期组胎盘血管内皮细胞中XIAP的表达(OD:0.18±0.05)显著低于正常对照组(OD:0.48±0.07),差异有显著性(t=19.097 9,P<0.01)。(3)绒毛血管密度在子痫前期的变化:子痫前期组胎盘绒毛血管密度[(56.14±6.22)个]明显低于对照组[(68.46±6.20)个](P<0.01),有统计学意义。结论子痫前期患者胎盘绒毛血管内皮细胞质中XIAP的表达明显下降,同时绒毛微血管密度降低,可能与血管内皮细胞凋亡增加导致子痫前期的发病有关。 Objective The purpose of this paper is to study the expression of anti-apoptosis protein XIAP in placental villi vascular endothelial cells and the density changes of villi vascular in preeclampsia. Methods Collected 60 pregnancies admitted into the Second Hospital of Shanxi Medical University and 30 pregnancies with preeclampsia is the study group while 30 normal pregnancies is the control group. Collected the clinical information and the childbirth placenta tissue of all enrolled pregnancies. The expression and the localization of XIAP in placental villi vascular endothelial cells of two groups were detected by immunohistochemical two footwork and the image was processed by Image-ProPlus medical image analysis system and then determined the OD for semi-quantitative analysis. HE stain was used to count villi vascular number. Statistical analyses were performed by two independent sample t test using SPSS 17.0. Results Comparison between two groups of clinical data: the age and gestational age of pregnancies in study group and control group had no significant differences (P〉0.05), while the systolic blood pressure, diastolic blood pressure, gestational age and birth weight in the two groups were markedly different (P〈0.05). Immunohistochemical results:XIAP positive expression can be found in the placental villi vascular endothelial cells of both study group and the expression quantity of XIAP in study group (OD:0.18±0.05) is significantly lower than control group (OD:0.48±0.07), there was a significant differences (t=19.097 9, P〈0.01). The density changes of villi vascular in preeclampsia:the density of villi vascular in study group (56.14±6.22) is significantly lower than control group (68.46±6.20), there was a significant differences. Conclusion Lower expression of XIAP in cytoplasm of placental villi vascular endothelial cells and decreased villi vascular may associate with the increasing apoptosis which caused preeclampsia.
