摘要
目的探讨Jagged1蛋白在早期自然流产患者绒毛组织中的表达及其对妊娠结局的影响。方法采用实时荧光定量逆转录聚合酶链反应技术和免疫印迹法检测15例自然流产患者和20例正常早期妊娠妇女绒毛Jagged1 mRNA和蛋白的表达水平。结果自然流产组患者Jagged1mRNA表达水平(0.062 3±0.000 7),低于正常早期妊娠妇女(0.031 5±0.0065),差异有统计学意义(P<0.05);Western-blot检测显示,早期自然流产患者Jagged1蛋白的表达水平低于正常早期妊娠妇女,但差异无统计学意义(P>0.05)。结论 Jagged1蛋白在早期自然流产患者绒毛组织中表达呈下调趋势,可能与母胎界面血管发育及滋养细胞缺氧有关,从而导致自然流产。
Objective To explore the expression of Jaggedl protein in villi of first trimester spontaneous abortion and its effects on pregnancy. Methods Real-time fluoreseenee quantitative RT-PCR and Western-blot were used to detect the expression levels of Jaggedl mRNA and protein in villi obtained from 15 women with first spontaneous abortion and 20 normal first trimester pregnant women. Results The expression levels of Jaggedl mRNA in villi of first trimester spontaneous abortion were significantly lower than those of the normal first trimester pregnancy( P 〈 0.05 ). Western-blot detection demonstrated that the expression levels of Jaggedl pro- tein in villi of the first trimester spontaneous abortion were lower than those of the normal pregnancy, hut without statistical significance ( P 〉 0.05 ). Conclusion The expression levels of Jaggedl protein in villi of the first trimester spontaneous abortion tended to be down- regulated, which might be associated with the vascular malformation and hypoxia in trophoblast at maternal-fetal interface, thus inducing spontaneous abortion.
