摘要
目的肌动蛋白相关蛋白2/3复合物5(ARPC5)参与癌细胞侵袭性伪足的形成,促进癌细胞的侵袭和转移。本研究旨在探讨肌动蛋白相关蛋白2/3复合物5(ARPC5)在非小细胞肺癌(NSCLC)细胞和组织中的表达特征及其与预后的相关性。方法 RT-PCR法及Western blot法检测ARPC5 m RNA及蛋白在肺癌细胞系A549、SK-MES-1、H460、95D及人正常支气管上皮细胞系HBE中的表达。免疫组化SP法检测ARPC5在133例NSCLC和25例癌旁正常组织中的表达情况,应用Kaplan-Meier曲线和Cox风险回归模型分析其与预后之间的关系。结果 ARPC5在NSCLC细胞系的m RNA及蛋白两个水平上的表达均高于正常支气管上皮细胞系。在133例NSCLC组织标本中,ARPC5阳性表达率67.7%,而在22例癌旁正常肺组织中不表达。ARPC5蛋白的表达与TNM分期和淋巴结转移密切相关(<0.01)。单因素生存分析显示ARPC5阴性表达患者5年生存率为57.8%,而阳性患2者为37.5%,差异有统计学意义(χ=8.634,<0.05);但多因素生存分析并未提示其为独立的预后指标。结论ARPC5是一种潜在的癌基因,可能参与肿瘤的转移,其表达影响患者预后但还不能成为判断肺癌预后独立指标。
Objective Actin related protein 2/3 complex 5(ARPC5) is involved in the formation of invadopodia and lamenllipodia, and plays a important role in metastasis and invasiveness of tumor cells. The aim of this study is to investigate the expressions and clinicopathologic significances of ARPC5 in non-small cell lung cancer(NSCLC), and to explore the its relationship with prognosis. Methods RT-PCR and Western blot were performed to detected the expression of ARPC5 m RNA and protein in lung cancer cell lines A549,SK-MES-1, H460,95 D and human normal bronchial epithelial cell line HBE. Immunohistochemistry(SP method) was performed to detect the expressions of ARPC5 in 133 NSCLC tissues and 25 paracancerous tissues. The Kaplan-Meier curve and the Cox proportional hazards model were used to evaluate the prognostic role of each studied parameter. Results Both of ARPC5 protein and m RNA were overexpressed in lung cancer cell lines, but negative in HBE. ARPC5 was detected in 133 NSCLC tissues and the positive rate of ARPC5 expression in NSCLC was 67.7%, with no expression of ARPC5 in 25 paracancerous tissues. The positive rate of ARPC5 was gradually increased with tumor progression, significantly higher in Ⅲ and Ⅳ stage cancers and in lymph node metastasis( 0.01), with no significant correlation with age, sex, T stage, histological type and differentiation grade(P〈0.05). P Univariate analysis showed that the 5-year survival rate of patients with negative ARPC5 expression was 57.8%, significantly 2higher than that of patients with positive ARPC5 expression(37.5%)(χ =8.634, P〈0.05). Multivariate Cox regression analysis showed that high expression of ARPC5 was not a significant independently prognostic factor. Conclusion ARPC5 is a potential oncogene and may be involved in metastasis of tumor. The expression of ARPC5 could affect prognosis of NSCLC, but still not a independently prognostic factor.
