人感染禽流感病毒A/Zhejiang/LS01/2014(H7N9)株HA基因序列分析

Sequence analysis of hemagglutinin gene of avian influenza virus A/Zhejiang/LS01/2014(H7N9) strain

摘要 目的测定本市首例人感染禽流感病毒A/Zhejiang/LS01/2014(H7N9)株HA基因序列,分析H7N9禽流感病毒HA基因特点。方法采用real-time PCR检测患者H7N9禽流感病毒核酸,RT-PCR扩增病毒HA基因后测定核苷酸序列。结果实验室确诊丽水首例人感染H7N9禽流感患者。测序得到H7N9禽流感病毒长度为1 683 bp的HA基因核苷酸全序。A/Zhejiang/LS01/2014(H7N9)与我国江浙沪分离株HA基因核苷酸和氨基酸序列相似性都比较高,分别为98.8%~99.5%、98.9%~99.5%;与浙江鸡分离株A/chicken/Zhejiang/C483/2013(H7N9)序列相似性最高。A/Zhejiang/LS01/2014(H7N9)与我国江浙沪H7N9病毒株HA基因序列均处于同一进化分支;与浙江外环境分离株A/environment/Hangzhou/34/2013(H7N9)进化关系最近;与浙江鸡分离株A/chicken/Zhejiang/C483/2013(H7N9)进化关系次之;与韩国野鸟分离株A/wildbird/Korea/A3/2011(H7N9)处于同一进化分支。结论 A/Zhejiang/LS01/2014(H7N9)HA基因来源于浙江鸡携带的H7亚型流感病毒。HA基因具有低致病性禽流感病毒基因特征,但与人类呼吸道上皮细胞的亲和力增加,更容易感染到人。 Objective To determine the first case of human infection with avian influenza virus hemagglutinin gene (HA) se- quences of A/Zhejiang/LSO1/2014(HTN9) strain in Lishui, and analyze the characteristics of HA gene of avian influenza virus HTN9. Methods Real - time PCR was used for the detection of human infection with H7N9 avian influenza virus nucleic acid, and the nucleotide sequence of HA gene was determined after amplified by RT - PCR. Results The first case of Lishui human infection with the H7N9 avian flu was confirmed in the laboratory. Sequencing and getting length of 1 683 bp HA gene nucleotidc sequence in HTN9 avian flu virus. The nucleotide and amino acid sequences of A/Zhejiang/LSO1/2014(HTN9) HA gene were highly similar with isolates of Jiangsu, Zhejiang and Shanghai, respectively, between 98.8% -99.5% and 98.9% -99.5% ; the highest sequence similarity existed with the separates of Zhejiang chicken strain A/chicken/Zhejiang/CA83/2013 (H7N9). A/Zhejiang/LS01/2014(H7Ng) strain of the H7N9 virus HA gene sequences and: the one from Jiangsu, Zhejiang and Shanghai were the same evolutionary branch; there was the closest evolutionary relationships with Zhejiang external environment isolates A/environment/Hangzhou/34/2013 (H7N9) ; followed by Zhejiang chicken isolates A/chicken/Zhejiang/C483/2013 (H7N9). All in the same evolutionary branch to South Korea wild isolates A/wildbird/Korea/A3/2011 (H7N9). Conclusion A/Zhe- jiang/LS01/2014(H7N9) HA gene origins from chickens carrying Zhejiang I-I7 subtype of influenza virus. HA gene with low pathogenic avian influenza virus genetic characteristics, but the affinity of human airway epithelial cells increases, more likely to infect humans.
