摘要 目的检测EPCAM、MUC16两种抗原在上皮性卵巢癌循环肿瘤细胞中表达的相关性。方法分别用羧基磁珠制备EPCAM、MUC16两种免疫磁珠,分3组:EPCAM免疫磁珠检测组、MUC16免疫磁珠检测组、2种磁珠均等混合检测组。对来源于同一患者的等体积外周血样本进行循环肿瘤细胞检测,HE染色后鉴定检测值。结果 EPCAM免疫磁珠、MUC16免疫磁珠、EPCAM+MUC16免疫磁珠对同体积、同来源样本检测值分别为22±11、14±6、28±12个肿瘤细胞(P<0.05),说明检测效率有统计学差异。结论 MUC16与EPCAM在上皮性卵巢癌循环肿瘤细胞中独立表达且存在差异,因此增加MUC16这一器官标志物可有效提高上皮性卵巢癌CTCs检出数量。 Tumor cells from primary fico shed into the bloodstream known as circulating tumor cells(CTCs),which is the important intermediate process of tumor metastasis. EPCAM(epithelial cell adhesion molecule) and MUC16(cell adhesion molecule 16) antigens were membrane markers of epithelial ovarian cancer cells which could be used to detect the target cells. This study performed to analyze the relation of EPCAM and MUC16 antigens expression in CTCs of ovarian cancer patients. Carboxyl beads labeled EPCAM and MUC16 antibodies were prepared, respectively. Immunomagnetic beads labeled EPCAM, MUC16 antibodies and mixtures of these two beads were spiked into three aliquots blood specimens to detect circulating tumor cells in patient. The detection rates were22±11, 14±6, 28±12(P〈0.05), suggesting that MUC16 and EPCAM expression are independent in circulating tumor cells of epithelial ovarian cancer, thus adding the marker of MUC16 could improve ovarian cancer detection rate of CTCs effectively.
