摘要
补体系统在先天免疫防御中起重要作用,但是过分激活补体会导致严重的组织伤害。C5a是补体激活的重要产物之一,参与败血症、急性肺损伤(acute lung injury,ALI)、过敏症及哮喘等许多炎症及自身免疫类疾病的发生与发展。使用抗C5a单克隆抗体结合C5a阻断该信号通路,可有效减轻炎症反应,为治疗炎症及自身免疫类疾病提供了新的思路。本文对C5a的结构、功能及其抗体药物的研究进展作一简要综述。
Complement system plays an important role in the innate immune defense. However, the excessive activation of complement may lead to serious tissue damage. As one of the important products of complement activation, CSa partici- pates in occurrence and development of inflammatory and autoimmune diseases, including sepsis, acute lung injury (ALI), allergy and asthma. Anti-CSa monoclonal antibody may neutralize CSa and block the signal pathway, thus relieves the inflammation reaction, which provides a new insight for the treatment of inflammatory and autoimmune diseases. The structure and function of CSa as well as the development of its antibody drugs are reviewed in this paper.
