摘要
目的:明确免疫鼠血中抗AT1受体自身抗体的类型分布,为该抗体的作用机制研究提供一定的实验依据。方法:以AT1R-ECII为抗原主动免疫雌性Wistar大鼠,待大鼠血清中含有高滴度的抗AT1受体自身抗体后,用ELISA法检测血清中抗AT1受体抗体的水平、类型及亚类。结果:雌性Wistar大鼠免疫4周后,抗AT1受体抗体滴度时明显升高,8周时处于稳定高峰值(免疫组OD值2.99±0.11 vs.同期对照组0.44±0.06,P<0.05);该抗体属于免疫球蛋白的Ig G类;在大鼠Ig G的4个亚型(Ig G1,Ig G2a,Ig G2b,Ig G2c)中,该抗体大部分属于Ig G2b亚类(OD值为0.33±0.04 vs.同期对照组OD值0.08±0.01,P<0.05),极少部分属于Ig G2a亚型(OD值为0.10±0.01 vs.同期对照组OD值0.07±0.05,P<0.05)。结论:通过主动免疫大鼠体内产生的抗AT1受体抗体主要属于Ig G类,Ig G2b亚类。
Objective: To detect distribution of the types of autoantibodies against AT1 receptor in the sera of immunized rats, providing the experimental basis for the research of the action mechanism. Methods: Adult female Wistar rats were actively immunized with AT1R-ECII. When the titer of anti-AT1R antibody reached a peak, the titers, types and subtypes of anti-AT1R antibody in the sera of rats were detected by SA-ELISA. Results: The titers of serum anti- AT1R antibody in immunized group were increased at the 4th week and attained a peak value at the 8th week as compared with the control group at the same time point ( OD value, 2.99 ±0.11 vs. 0.44 ± 0.06, P 〈 0.05 ). It was shown that anti-AT1R antibody was IgG type immunoglobulin( IgG1, IgG2a, IgG2b, IgG2c). Meanwhile, the antibody was mainly determined as IgG2b subtype ( OD value, 0. 33 ± 0.04 vs. 0.08 ± 0.01, P 〈 0.05 ), and a few belonged to IgG2a subtype( OD value, 0.10± 0.01 vs. 0.07 ±0.01, P 〈 0.05 ). Conclusion :The anti-AT1 R antibodies in immunized rats aremainly determined as IgG2b subtype, and a few belongs to IgG2a subtype.
