摘要
目的探讨丹红水溶性有效组分配伍对缺血后再灌注损伤心肌保护作用。方法 SD大鼠冠脉结扎40min造成心肌缺血后剪开丝线再灌注120 min建立心肌缺血后再灌注损伤实验模型,其中10 min后股静脉注射丹参水溶性有效组分作为A组,注射红花水溶性有效组分作为B组,丹参红花水溶性有效组分配伍作为C组,而注射等量生理盐水作为对照组,每组10只,测定心肌梗死面积、血清肌酸激酶同工酶(CK-MB)、肌钙蛋白T(c Tn T)、心功能指标[左室收缩期平均压(LVSP)、舒张末期压力(LVEDP)和射血分数(EF)],采用放射免疫法检测血浆降钙素基因相关肽(CGRP)、6-酮基-前列腺素F1α水平的差异。结果与对照组比较,A、B、C三组心肌梗死面积减少,CK-MB、c Tn T和LVEDP均明显降低,而LVSP、EF、CGRP和6-酮基-前列腺素F1α明显增高,组间比较差异具有统计学意义(P<0.05),其中C组心肌梗死面积最小,CK-MB、c Tn T、LVSP、EF、LVEDP、CGRP和6-酮基-前列腺素F1α改善幅度最大,与A、B两组比较差异具有显著性(P<0.05),而A、B组上述指标改善幅度比较差异无显著性(P>0.05)。结论丹红水溶性有效组分配伍通过减少心肌梗死面积、降低心肌酶水平和升高CGRP、6-酮基-前列腺素F1α水平而起到保护缺血再灌注损伤心肌、改善心功能的作用。
Objective To investigate safflower water soluble effective components compatibility in myocardial protective effect for rat is-chemia reperfusion injury. Methods SD rats with 40 min myocardial ischemia caused by coronary artery ligation and cut open the thread 120 min of reperfusion after myocardial ischemia reperfusion injury were established as experimental model,in which 10 min after intravenous injection of Salvia miltiorrhiza water-soluble effective components were considered as A group,safflower injection water soluble effective components were considered as B group,Salvia miltiorrhiza safflower water soluble effective components compatibility were considered as the C group,and normal saline injection were considered as control group,10 rats in each group. Serum creatine kinase myocardial infarction area and isoenzyme( CK-MB),cardiac troponin T( cTnT)and heart function index left ventricular systolic pressure( LVSP),diastolic pressure( LVEDP)and ejection fraction( EF)were detected. Plasma calcitonin gene related peptide by radioimmunoassay( CGRP),6- keto prostaglandin F1αwere were detec-ted by radioimmunoassay. Results Compared with control group,myocardial infarct size,CK-MB,cTnT and LVEDP were significantly de-creased,while LVSP,EF,CGRP and 6- keto prostaglandin F1 α of A,B,C groups were reduced significantly higher,which had significant difference was statistically( P 〈0. 05). Myocardial infarct area of C group was the least. CK-MB,cTnT,LVSP,EF,LVEDP,CGRP and 6-keto prostaglandin F1αsignificantly improved,and it had significant difference between the A,B two groups( P 〈0. 05),and there was no signif-icant difference in the parameters mentioned above in group A,B groups( P 〉0. 05). Conclusion Salvia miltiorrhiza safflower water soluble ef-fective components compatibility reduces myocardial infarction area,myocardial enzyme level and increased CGRP and 6 - keto prostaglandin F1αlevel to achieve protecting myocardial ischemia reperfusion injury,improve heart function.
