白藜芦醇通过调控Wnt/β-catenin信号通路抑制结肠癌细胞生长的研究 被引量:5

摘要 目地探讨白藜芦醇抗肿瘤活性的作用机制。方法本研究使用白藜芦醇处理体外培养结肠癌细胞系HCT116后,通过细胞生长实验和MTT增殖实验来探讨其对HCT116细胞生长、增殖的影响;通过免疫印迹实验检测β-catenin的表达水平和Wnt信号通路靶基因c-Myc和cyclinD1的表达,检测白藜芦醇对Wnt信号通路活化的影响。结果白藜芦醇能显著抑制HCT116的生长增殖,降低β-catenin的表达水平,抑制Wnt信号通路靶基因c-Myc和cyclinD1的表达。结论白藜芦醇通过抑制Wnt信号通路从而抑制肿瘤生长,本研究为白藜芦醇抗肿瘤机制提供了新的思路和方向。 Objective To investigate the anti-tumor mechanism of resveratrol. Methods Colon cancer cell lines HCT116 were treated with resveratrol, and cell growth curve and MTT proliferation were performed. The expression of β-catenin and Wnt signaling pathway target genes c-Myc and cyclinD1 were detected by immunoblotting. Results Resveratrol significantly inhibited the growth and proliferation of HCT116, and reduced the expression levels of β-catenin, c-Myc and cyclinD1. Conclusion Resveratrol inhibits tumor growth through blocking Wnt signaling pathway, which provides a new direction for the anti-tumor mechanism of resveratrol.
