蒙医灸疗对失眠模型小鼠脑组织一氧化氮和一氧化氮合酶的影响

Effect of moxibustion of Mongolian medicine on nitricoxide and nitric oxide synthase in brain tissue of mice model of insomnia

目的探讨施灸"赫依穴""顶会穴"对睡眠紊乱模型小鼠脑组织中一氧化氮(NO)和一氧化氮合酶(NOS)的影响。方法将36只小白鼠随机分为空白对照组、模型组、治疗组3组,每组12只,模型组与对照组反复定时腹腔内注射咖啡因致小鼠睡眠节律紊乱,治疗组施灸"赫依穴"与"顶会穴"。观察各组小鼠自发活动次数与站立数,检测小鼠脑组织中NO含量与NOS活性。结果各组小鼠站立次数与自主活动次数比较:治疗组与空白对照组之间无统计学意义(P>0.05);与模型组比较空白组和治疗组均有统计学意义(P<0.05)。各组小鼠脑组织中NO含量和NOS活性比较:治疗组与空白对照组之间无统计学意义(P>0.05);与模型组比较,空白对照组和治疗组均有统计学意义(P<0.05)。结论蒙医灸疗对睡眠紊乱模型小鼠具有镇静催眠作用,其机制可能是通过阻抑NOS诱导产生过量的NO起到神经保护作用。

Objective To investigate the Moxibustion ＂point, Heyi＂＂point, Dinghui＂ on sleep disorder mice brain tissues of NO and NOS effect. Methods The 36 mice were randomly divided into blank control group, model group, treatment group three groups, 12 rats in each group,model group and control group repeated intraperitoneal injection of caffeine in mice in- duced by the timing of sleep rhythm disorder, the treatment group Moxibustion ＂Heyi point＂ and ＂Dinghui point＂. The mice were observed the times of spontaneous activities and stand number, NO content and NOS activity in brain tissue of mice were detected in. Results compared mice the number of standing and independent activity times： there is no significant dif- ference between treatment group and blank control group （ P 〉 0.05）, compared with the model group, the blank group and the treatment groups were significantly different （ P 〈 0.05） ; the comparison of NO content and NOS activity in brain tissue of mice in each group： there was no significant difference in between treatment group and blank control group （ P 〉 0.05）, compared with the model group, blank control group and the treatment groups were significantly different （ P 〈 0.05）. Con- clusion Moxibustion of Mongolian medicine has sedative and hypnotic effects on sleep disturbance model mice, may be in- duced to produce excessive NO by inhibiting NOS, play a neuroprotective effect.
