干扰素诱导基因AIM2与系统性红斑狼疮的相关性研究 被引量:3

摘要 目的:探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)中干扰素诱导基因AIM2的表达及其与疾病活动度的相关性。方法:运用半定量逆转录聚合酶链反应(RT-PCR)技术检测30例SLE患者(SLE组)与25例健康体检者(正常对照组)的AIM2 m RNA表达水平,并且分析它们与尿蛋白、补体C3、补体C4、抗ds-DNA抗体及SLE疾病活动指数(SLEDAI积分)的相关性。结果:1与正常对照组比较,SLE组AIM2 m RNA表达显著增高(P<0.01);2 SLE组尿蛋白阳性患者的AIM2 m RNA表达水平较尿蛋白阴性患者及正常对照组显著增高(P<0.01),尿蛋白阴性组AIM2 m RNA表达水平与正常对照组比较,差异无统计学意义(P>0.05);3 SLE组AIM2 m RNA表达水平与补体C3呈显著负相关(r=-0.623,P<0.05),与抗ds-DNA抗体、SLEDAI积分呈显著正相关(r=0.621、r=0.503,P<0.05),与补体C4不相关(r=-0.426,P>0.05)。结论:SLE患者AIM2 m RNA的表达水平显著增高,并与肾脏损伤、疾病活动度相关。 Objective:To investigate the relationship between the expression of interferon-induced gene AIM2 in peripheral blood mononuclear cells(PBMCs) of patients with systemic lupus erythematosus(SLE)and the disease activity.Methods:Semi-quantitative Reverse Transcription Polymerase Chain Reaction(RTPCR) was used to detect the expression levels of AIM2 mRNA of 30 patients with SLE(the SLE group) and 25 healthy person(the normal control group),analyzing their relationship with urinary protein,complements C3 and C4,anti ds-DNA antibody and SLE disease activity index(SLEDAI integral).Results:① Compared with the normal control group,the expression of AIM2 mRNA in the SLE group increased significantly(P〈0.01);② The expressions of AIM2 mRNA in patients with positive urinary protein in the SLE group were significantly higher(P〈0.01) than those with negative urinary protein in the group and all in the normal control group.The expressions of AIM2 mRNA in patients with negative urinary protein had no statistical significance(P〉0.05)from those of the normal control group. ③ The expression levels of AIM2 mRNA in the SLE group had a significant negative correlation with complement C3(r =- 0.623,P〈0.05), an obvious positive correlation with anti ds-DNA antibody and SLEDAI integral(r = 0.621,r = 0.503,P〈0.05) and no relationship with complement C4(r =- 0.426,P〉0.05).Conclusion:The expression levels of AIM2 mRNA in SLE patients significantly increase,which are associated with kidney injury and disease activity.
