摘要
目的探讨Src激酶特异性抑制剂PP2对人胆管癌QBC939细胞侵袭能力的影响和机制。方法通过Western Blot技术检测PP2对人胆管癌QBC939细胞中Src激酶活化的影响;用Transwell小室法观察PP2对QBC939细胞的影响;用RT-PCR和Western Blot技术检测PP2对QBC939细胞侵袭能力相关分子的作用。结果 PP2组中p-Src蛋白的表达水平明显低于对照组;PP2组QBC939细胞的体外侵袭能力较对照组显著降低;与对照组相比,PP2组E-cadherin的表达显著增强,CD44的表达则明显减弱。结论 PP2能通过抑制Src激酶的活化,增强QBC939细胞侵袭抑制分子E-cadherin、减弱促侵袭分子CD44的表达,进而抑制人胆管癌QBC939细胞的侵袭能力。
[ Objective ] To detect the effect of Src kinase inhibitor PP2 on the invasion of human cholangiocarci- noma QBC939 cells. [ Methods ] Western blot was used to detect the effect of Src kinase inhibitor PP2 on Src acti- vation. Transwell chamber assay was performed to detect the effect of Src kinase inhibitor PP2 on the invasion of hu- man eholangioearcinoma QBC939 cells. The effect of Src kinase inhibitor PP2 on the invasion-related molecules of human cholangioearcinoma QBC939 cells was detected by RT-PCR and western blotting. [ Results ] The expression of p-Src protein in PP2 group was significantly lower than that in control group. The invasion of QBC939 cell in PP2 group was obviously decreased than that in control group. Compared with control group, E-cadherin expression in PP2 group was observably enhanced, and CD44 expression was dramaticlly decreased. [ Conclusions ] PP2 can in- hibit Src kinase activation, increase the expression of E-cadherin, reduce the expression of CD44, then inhibit the invasion of human eholangiocarcinoma QBC939 cells.
