摘要
目的 比较妊娠小鼠与未孕小鼠乳腺上皮细胞中微小RNA(miRNA,miR)的表达差异,探讨早孕降低乳腺癌风险的机制.方法 使用miRNA基大芯片筛选出已孕和未孕小鼠乳腺上皮细胞中差异表达的miRNAs,从中挑选变化幅度较明显的miR-132分析.然后,验证miR-132对小鼠乳腺癌细胞增殖、侵袭能力的影响.结果 芯片结果提示,和未孕小鼠比较,已孕鼠的乳腺上皮细胞中miR-132表达水平上调约2.6倍,差异有统计学意义(P<0.05).在小鼠4T1乳腺癌细胞中过表达miR-132之后,可以抑制小鼠乳腺癌细胞的增殖(下降2.3倍)及侵袭能力(下降2.1倍).结论 已孕和未孕鼠乳腺上皮细胞中miRNAs的表达差异性,可能与乳腺癌的患病风险相关.
Objective The aim of this article is to explore the mechanism of early pregnancy in reducing the risk of breast cancer through comparing the microRNAs (miRNA,miR) differences between pregnant and virgin mice.Methods The miRNAs differences between pregnant and virgin mice were chose with the use of microrna array.We picked up miR-132 for further study.We investigated the miR-132 effect on proliferation and invasion in mice breast cancer cells.Results miR-132 expression was elevated approximately 2.6 folds in pregnant mice when compared with virgin mice.Overexpression of miR-132 inhibited cell proliferation ability (2.3 folds) and invasion ability (2.1 folds) in mice breast cancer cells.Conclusion The miRNAs differences between pregnant and virgin mice may be associated with breast cancer risk.
