摘要
目的:观察大承气汤对不完全性肠梗阻大鼠血清中内毒素(ET)和肿瘤坏死因子(TNF-a)含量变化的影响。方法:将60只SD大鼠根据随机分组法分为6组:空白对照组,模型对照组,假手术组,大承气汤高、中、低剂量组,每组10只。对除空白对照组和假手术组外的其余组大鼠制备不完全性肠梗阻模型,假手术组采用针线只穿透肠系膜而不结扎的方法处理。大承气汤高、中、低剂量组分别按1.16、0.58、0.29g/m L的剂量给予大承气汤灌胃治疗,其余3组给予相同剂量的生理盐水灌胃。运用酶联免疫法(ELISA)检测各组大鼠血清中ET、TNF-α的表达量,并进行比较。结果:与模型对照组比较,大承气汤高、中剂量组血清中ET含量及TNF-α含量均明显降低,差异有统计学意义(P<0.01)。结论:大承气汤能够降低不完全性肠梗阻大鼠血清ET及TNF-a含量,保护不完全性肠梗阻小肠上皮细胞,具有肠屏障保护功能。
Objective: To observe the effects of Dachengqi decoction on the serum content of endotoxin( ET)and tumor necrosis factor( TNF)- in rats with incomplete intestinal obstruction. Methods: Sixty Sprague- Dawley rats were equally and randomly divided into 6 groups: blank control group,model control group,sham- operation group,and high-,middle-,and low- dose Dachengqi decoction groups. A rat model of incomplete intestinal obstruction was established in all the groups except the blank control group and sham- operation group. The mesenterium of the rats in the sham- operation group was threaded through with a needle and thread but without ligation. The rats in the high-,middle-,and low- dose Dachengqi decoction groups were treated by intragastric administration of Dachengqi decoction at 1. 16,0. 58,and 0. 29 g / m L,respectively. The remaining three groups were intragastrically administered with normal saline at the same dose. The serum content of ET and TNF- in the rats of each group was determined by enzyme- linked immunosorbent assay and compared. Results: Compared with that in the model control group,the serum content of ET and TNF- a in the high- and middle- dose Dachengqi decoction groups significantly decreased( P〈0. 01). Conclusion: Dachengqi decoction can reduce the serum content of ET and TNF- in rats with incomplete intestinal obstruction,which protects small intestinal epithelial cells and has intestinal barrier function for patients with incomplete intestinal obstruction.
