补肾活血颗粒对帕金森病模型大鼠黑质和纹状体中Fas、FADD表达的影响 被引量:6

Influence of Bushen Huoxue Granule( BSHXG) on the expression of Fas and FADD of substantia nigra and striatum in model rats with Parkinson's disease

摘要 目的探讨补肾活血颗粒对帕金森病模型大鼠黑质、纹状体中Fas、FADD表达的影响。方法 60只雄性SD大鼠随机分为正常组10只和造模组50只,采用6-羟基多巴胺注射造模。造模成功的23只随机分为模型组10只和观察组13只。观察组给予补肾活血颗粒灌胃,正常组和模型组给予等量生理盐水灌胃。应用免疫组化染色技术观察大鼠黑质、纹状体中Fas和Fas死亡结构域蛋白(FADD)表达的变化情况。结果模型组Fas在黑质阳性细胞表达率(PI)为(1.400±0.070),纹状体PI为(1.820±0.090),均高于正常组和观察组(P<0.01)。模型组FADD在黑质PI为(1.160±0.058),纹状体PI为(0.780±0.040),均高于正常组和观察组(P<0.01)。结论补肾活血颗粒治疗帕金森病的疗效机制可能与下调Fas和FADD表达有关。 Objective To explore the influence of Bushen Huoxue Granule (BSHXG) on the expression of Fas and Fas-assoeiated death domain (FADD) levels in substantia nigra and striatum of model rats with Parkinson' s disease (PD). Methods 60 male SD rats were randomized into a normal control group (10 rats) and a model group (50 rats) which were induced by injecting 6- OHDA. Then, 23 successful model rats were randomized into a model group (10 rats) and an observation group (13 rats). The observation group was treated by Bushen Huoxue Granule with intragastric administration, the normal group and the model group were given the same amount of saline with intragastric administration. Immunohistochemical staining was used to observe the expression changes of Fas and FADD in substantia nigra and striatum of rats. Results Fas expression positive ceils(PI) in the substantia nigra and the striatum of the PD model group were 1. 400 ± 0. 070 and 1. 820 ± 0. 090 respectively, higher than that of the normal group and the observation group (P 〈 0. 01 ) . FADD PI values in the substantia nigra and the striatum of the PD model group were 1. 160 ±0. 058 and 0. 780 ± 0. 040 respectively, higher than that of the normal group and observation group (P 〈 0. 01 ) . Conclusion The therapeutic effect of BSHXG on PD is possibly attributed to the decreased content of Fas and FADD.
