摘要
目的建立一种快速测定人尿中硫氰酸盐的方法。方法采用三氯化铁分光光度法,准确吸取5.00 ml样液于10 ml带塞比色管中,然后加入0.2 ml三氯化铁溶液,加塞摇匀放置10 min后,用1 cm比色杯,纯水调零,在波长465 nm处测定吸光度。结果对3份不同试样按本方法的试验操作条件,6 d内重复测定6次,3份不同试样的平均值(x)分别为1.23、8.60、12.60;标准偏差(s)分别为0.02、0.03、0.04;变异系数(CV)分别为1.63%、0.35%、0.32%;在3份不同试样中分别加入不同量的硫氰酸盐标准溶液,分别测定,3份不同试样的回收率分别为92.2%、92.7%、94.3%。结论用该方法测定人尿中硫氰酸盐的含量,其精密度试验和回收率试验结果令人满意。
Objective To establish a method for rapid determination of thiocyanate in human urine. Methods Ferric triehlo- ride was used to determine thioeyanate by speetrophotometry, 0.2 ml ferric chloride solution was added to 5.00 ml sample in 10 ml color tube, shaken up with the solution and placed for 10 rain. A euvette with lem width was used in this work, and cor- rected zero by pure water with the determination wavelength of 465 nm. Results According to the method of experimental con- ditions, 3 different samples were constant determined for 6 times in 6 days, the sample average value were respectively 1.23, 8.60, 12.60: the standard deviations (s) were respeetively 0.02, 0.03, 0.04; the coefficients of variation (CV) were respec- tively 1.63%, 0. 35%, 0. 32% ; the different amounts of thioeyanate standard solution were added in 3 different samples which were determined respectively. The recovery rates of the different samples were respectively 92.2%, 92.7%, 94.3%. Conclusion The method was successfully applied to determine thiocyanate in human urine, and the precision and recovery results were satisfactory.
