摘要
目的探讨多重耐药鲍曼不动杆菌外排泵蛋白在耐药方面的作用和外排泵基因表达情况。方法分离自河北医科大学第一医院的多重耐药鲍曼不动杆菌,加入外排泵抑制剂后MIC值降低至原值的1/4或以上判定为外排泵表型阳性株,外排泵表型阳性鲍曼不动杆菌34株采用PCR扩增法对其进行外排泵蛋白基因ade B的检测,并将ade B基因测序序列与Gen Bank中序列进行同源性比较。结果 34株外排泵表型阳性的鲍曼不动杆菌检测到ade B基因的有33株,检出阳性率为97.06%。对33株鲍曼不动杆菌的外排泵基因ade B进行测序,经比对,所测序列与Gen Bank中序列同源性为100%。结论 Ade ABC外排泵机制在鲍曼不动杆菌对抗菌药物敏感性方面的作用尤其重要,含有ade B基底泵的重组衍生体相对于不含有的菌株抗菌素的敏感性降低,所以主动外排基因ade B是鲍曼不动杆菌发生多重耐药的重要因素之一。
Objective To investigate the resistant function and efflux pump gene expression of the effiux pump protein of muhi- pledrug resistant Acinetobacter baumannii. Methods 34 strains multipledrug resistant Acinetobacter baumannii separated from the first hospital of Hebe medical universityi were positive efflux pump phenotype, after added into efflux pump inhibitors, MIC value reduced to a quarter or more of the original value, were detected as the efflux pump phenotypic positive strains. The efflux pump protein gene adeB of 34 strains of effiux pump positive Acinetobacter baumannii was detected by PCR amplification method and compared adeB gene sequence with those homologolus sequence in GenBank. Results Among the 34 stains efflux pump phenotype positive Acinetobacter baumannii, there were 33 strains adeB gene, and the positive rate was 97.06%. To sequence 33 strains Acinetobacter baumannii adeB efflux pump gene, after compared these sequences, the sequence homology was 100% in GenBank. Conclusion AdeABC efflux pump mechanism is especially important in Acinetobacter baumannii antimicrobial sensitivity, the sensitivity of derivatives containing adeB gene reduced. The active efflux pump adeB gene plays a important role in Acinetobacter baumannii muhipledrug resistance.
