摘要
将硫酸阿米卡星与大肠埃希菌血清抗体用化学修饰剂进行偶联,再经化学方法处理,按照制剂学原理制备成靶向阿米卡星。并对在不同环境下的偶联物进行革兰染色,以原制剂做对照,做体外抑菌试验,把检测结果与染色涂片做比较,探索革兰染色在靶向制剂稳定性和有效性的简单应用。结果表明,当不同环境下靶向阿米卡星的革兰染色均与原制剂染色结果相同时,其与原制剂的MIC与MBC结果也相同,若染色结果不同则MIC与MBC都有不同程度的减弱或抑菌效果消失,革兰染色结果可以简单观察靶向阿米卡星的稳定性和有效性。
Through coupling of amikacin sulfate and serum antibody against Escherichia coli by chemical modifiers and chemi- cal treatment, the preparation of targeting amikacin can be achieved according to principles of pharmaceutics. At the same time, gram stain was made for conjugates in different environment and nti-bacterial experiments were carried out ain vitro by comparison with original preparations, and through comparing testing result and stain smear, the stability and effectiveness of simple applica- tion of gram stain in targeting drug delivery system were explored. We found that the results of amikacin-targeting gram stain in dif- ferent environments were all identical to the staining results of original preparations, the MIC and MBC were also the same as those of original preparations ; if the staining results were different, and then the MIC and MBC would be weakened at varying degrees or the antibacterial effect would disappear. The result of gram stain contributed to simple observation of the stability and effectiveness of the targeted amikacin.
