基于化学显色反应的黄菠萝雌雄株鉴别技术研究 被引量:1

摘要 为研究黄菠萝幼苗早期雌雄鉴定技术,采用甲基红法、溴麝香草酚蓝(BTB)法和氯化三苯基四氮唑(TTC)法和斐林试剂等化学药剂对黄菠萝叶片提取液进行染色,同时紫外分光光度计测定光谱吸收值,通过成熟黄菠萝雌雄株叶片提取液颜色和光谱值的差异,推断黄菠萝幼苗性别。结果表明,甲基红溶液染色的提取液在30℃水浴锅中3 h颜色差异显著,雄性植株提取液呈现棕红色,雌性植株提取液为桔红色;BTB法雌雄株提取液颜色差异明显,雄性植株提取液颜色呈深蓝色,雌性植株提取液为棕黄色,吸收光谱值差异显著(P<0.01)。TTC法在485nm下测定雌雄株提取液存在显著差异(P<0.01)。斐林试剂的显色反应和雌雄株提取液吸收光谱值存在差异(0.01<P<0.05),表明甲基红法、BTB法、TTC法和斐林试剂法均可作为黄菠萝雌雄株鉴定的方法。 In order to study the early gender identification techniques for Phellodendron amurense, methyl red, BTB, TTC and Fehling's solution were used to stain the extract of Phellodendron amurense, and the UV spectrophotometer value was obtained by using ultraviolet spectrophotometer. The different color of extract and spectral value of Phellodendron amurense were used to infer the gender of sapling. The results showed that the chromogenic reaction was significantly different at 30 ℃ water bath for three hours, the solution of male extract showed brown and red color, while the color of the solution of female extract was orange; With BTB method, the differences of the plant extracts were significant, the male plant extract color was dark blue, and the female plant extract color was brown. The absorption spectra values were significantly different(P 〈0. 01 ). With TTC method measured at 485nm, the differences of the male and female plant extracts were significantly different(P 〈0. 01 ). The color reaction of Fehling's solution for male and fe- male plant extracts and the spectral values were significantly different( 0. 01 〈 P 〈 0. 05 ), which showed that methyl red method, BTB method, TTC method and Fehling's solution methods can be used to identify the gender of Phellodendron amurense.
