复元活血汤加味改良对大鼠慢性高眼压模型视神经损害的保护作用及分子机制探究 被引量:5

摘要 目的:研究复元活血汤加味改良对大鼠慢性高眼压模型视神经损害的保护作用及分子机制。方法:选择SD大鼠作为研究对象,随机分为正常组、模型组以及治疗组,采用巩膜静脉烧灼法制作高眼压大鼠模型,给予复元活血汤球后注射进行治疗,治疗后检测视网膜中凋亡基因、细胞因子、水通道和离子通道的含量。结果:(1)凋亡基因:与模型组比较,治疗组大鼠视网膜中c-fos、c-jun、P53、Bax、Caspase-3以及Smac的mRNA含量较低(P<0.05);(2)细胞因子:与模型组比较,治疗组大鼠视网膜中脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)、神经营养素3(neurotrophin-3,NT-3)、神经调节蛋白1(neuregulin-1,NRG-1)、胰岛素样生长因子-1(insulin-like growth factor 1,IGF-1)的mRNA含量较高,基质细胞衍生因子-1(SDF-1)、碱性成纤维细胞生长因子(b-FGF)含量较低(P<0.05);(3)水通道和离子通道:与模型组比较,治疗组大鼠视网膜中水通道蛋白(AQP)-1和AQP-4的mRNA含量较高,电压门控氯离子通道(CIC-3)、PV、CB和CR的mRNA含量较低(P<0.05)。结论:复元活血汤治疗有助于抑制视网膜神经节细胞凋亡,调节细胞因子的生成以及水通道,离子通道的表达,对大鼠慢性高眼压模型视神经损害具有保护作用。 Objective: To study the protective effect and molecular mechanisms of modified Fuyuan Huoxue decoction on optic nerve damage in chronic high intraocular pressure model of rat. Methods.. SD rats were selected as research subjects and randomly divided into normal group, model group, and treatment group. High intraocular pressure models of rats were established through episcleral vein cauterization; retrobulbar injection of Fuyuan Huoxue decoction was conducted for treatment. Then contents of apoptotic genes, cytokines, water channels and ion channels in retina were detected after treatment. Results: Compared with model group, mRNA contents of c-los, c jun, P53, Bax, Caspase 3 and Smac, contents of SDF-1 and b-FGF, rnRNA contents of CIC-3, PV, CB and CR in retina of treatment group were lower; mRNA contents of BDNF, NT-3, NRG-1 and IGF-1, mRNA contents of AQP-1 and AQP-4 in retina of treatment group were higher. Conclusion; Fuyuan Huoxue decoction treatment is helpful to inhibit apoptosis of retinal ganglion cells and regulate generation of cytokines as well as expressions of water channels and ion channels; it has protective effect on optic nerve damage of chronic high intraocular pressure models of rats.
