摘要
目的探索中药补肾益脑片抗衰老的分子机制。方法使用DDRT-PCR方法研究中药补肾益脑片对加速衰老(SAMR1)小鼠脑组织mRNA表达的影响,观察SAMR1小鼠脑组织基因表达情况。结果发现并克隆了8个受补肾益脑片影响而表达下调的基因片段,分别编码为60S核糖体L21蛋白、164k Da中心体蛋白、泛素特异性蛋白酶14、2型神经营养性酪氨酸受体激酶、海马丰富基因转录物1、Bptf转录因子、X-染色体连锁α地贫/智力低下综合征蛋白和细胞色素C氧化酶亚单位Ⅱ;3个受补肾益脑片影响而表达上调的基因片段,分别编码为线粒体反义RNA、生长激素诱导膜蛋白和葡萄糖-果糖氧化还原酶。这些表达片段多数与已知的衰老相关基因同源。它们在体内涉及能量代谢、信号传导、蛋白质合成与降解、RNA合成与稳定和染色质的重塑等多个方面。结论补肾益脑片以多基因、多靶点、多效应方式发挥其抗衰老作用。
Objective It is to explore the anti-senescence molecular mechanism of the traditional Chinese medicine Bushen Yinao tablet. Methods DDRT - PCR was applied to compare the cerebral tissue gene expression of the Senescence-Accelerated Mouse R1 ( SAMR1 ) which were treated with traditional Chinese medicine Bushen Yinao tablet. Results In the TCM given group, we cloned 8 up-expression gene fractions which were 60S ribosomal protein L21, centrosomal protein 164kDa (CEP164) , ubiquitin specific protease 14, neurotrophic tyrosine kinase receptor type 2 (Ntrk2) , hippocampus abundant gene transcript 1 (Hiatl) , bromodomain PHD finger transcription factor (Bptf) , alpha thalassemia/ mental retardation syndrome X-linked homolog (human) (Atrx) , cytochrome c oxidase subunit Ⅱ (Cox2) , and 3 down-expression gene fractions which were Mitochondrial antisense RNA, growth hormone-inducible membrane protein mRNA, glucose-fructose oxidoreductase domain containing 1 ( Gfod1 ). Most of those were homologous to the known senescence genes. In organism, they involved in energy metabolism, signal transduetion, protein synthesis and Degradation, RNA Transcription and stabilization, and chromatin state remodel, etc. Conclusion This research demonstrated that Bushen Yinao tablet produced its effect with multigene, multiple targets and plurivalent effects in anti-senescence.
