摘要
建立测定原料奶中庆大霉素C组分的高效液相色谱法。通过加入适量沉淀剂除去样品中大部分蛋白质,采用邻苯二甲醛试剂柱前衍生化,利用C18色谱柱,以含有0.02 mol/L的甲醇-醋酸溶液为流动相进行梯度洗脱,用荧光检测器检测,激发波长340 nm、发射波长450 nm。结果表明,回收率在85.6%~97.3%,精密度(RSD)为1.21%~4.04%,最低检出限为0.2μg/m L。该方法可用于原料奶中庆大霉素残留4种组分的测定。
To establish a HPLC method for determination of gentamicin in the raw milk. Protein in milk were mostly eliminated by the precipitators, OPA was used for derivatization, and then separated on C 18 column with a mixed solution of methanol-acetic acid as mobile phase, and excitation and emission wavelength are 340 nm and 450 nm with a fluorescence detector The recovery were 85.6 %-97.3 % ,the relative standard deviations (RSD) were 1.21%-4.04 %, the minimum detectable values were 0.2μg/mL. This method can be used for the routine analysis of gentamicin in raw milk.
