叶酸修饰的重组Apoptin对胃癌治疗作用的实验研究

摘要 目的研究叶酸修饰的重组Apoptin(以下简称rApoptin)在体内外对胃癌的治疗作用。方法体外采用MTT法测定rApoptin对胃癌细胞生长抑制率;体内用胃癌荷瘤裸鼠研究rApoptin对肿瘤体积增长抑制作用及对裸鼠体重的影响。结果体外实验证明rApoptin对胃癌细胞有显著的生长抑制作用,并有明显的量效关系;对体内肿瘤体积亦有显著的抑制作用(P<0.01),病理结果显示,与对照组比较,实验组的肿瘤组织细胞发生大面积坏死或凋亡。结论 rApoptin胃癌组织细胞具有很强的杀伤作用。 Objective To investigate the effect of rApoptin on human gastric carcinoma BGC823 cells and xenograft growth in nude mice.Methods MTT(methyl thiszolyl tetrazolium)assay was used to determine the inhibition of BGC823 cells proliferation by rApoptin.BALB/c nude mice were used to determine antitumor activity in vivo.ResultsProliferation of BGC823 cells was inhibited in a dose-dependent manner in vitro and the CI50 was 6.2μg/ml.rApoptin could suppressed the growth of BGC823 xenografts in nude mice by 55% and 60% at 1.5mg/kg and 2.5mg/kg,respectively.HE staining showed most of the tumor cells were died or apoptosis.Conclusion rApoptin could inhibit the growth of human gastric carcinoma BGC823 cells in vitro and in vivo effectively.
