摘要 目的探讨miR-514在肾细胞癌中的表达变化及其临床意义。方法提取45例肾癌及其癌旁正常组织中的总RNA,逆转录后用实时荧光定量PCR的方法检测miR-514在肾癌组织和癌旁正常组织中的表达情况,并分析其与肾癌患者临床病理特征的关系。结果与相应癌旁组织相比,miR-514在肾癌组织中表达量显著下调,下调率为86.67%。miR-514表达水平与患者年龄、性别、病理分型、TNM分期没有关系,但其表达水平与Fuhrman分级有关。结论 miR-514可能与肾癌的发生发展有密切关系,进行miR-514表达水平的研究可能对肾癌的早期诊断及治疗具有潜在的临床意义。 Objective To investigate the expression levels of miR-514 in renal cell carcinoma and its clinical significance.Methods Total RNAs were isolated from 45 pairs of human renal cell carcinoma and matched adjacent normal tissues.After reverse transcription,the expression of miR-514 was analyzed by quantitative real-time PCR.Then the relationship between the expression level of miR-514 and the clinicopathological characteristics was analyzed.ResultsCompared with the corresponding adjacent tissues,the expression of miR-514 in renal cell carcinoma was significantly reduced,with an average of 4.14 fold down-regulation.There's no relation between the expression of miR-514 and the age,gender,pathological classification and TNM stage of patients with renal cell carcinoma.However,the expression level of miR-514 was related with the Fuhrman grade.Conclusion miR-514 may be involved in the development and progression of renal cell carcinoma.This study of miR-514 expression may present a potential significance in the early diagnosis and treatment of renal cell carcinoma.
