填埋场中多酚氧化酶活性测定条件优化 被引量:1

摘要 以填埋场垃圾为研究对象,在土壤酶测定方法为基础上,对垃圾降解过程中多酚氧化酶活性测定方法进行优化。通过单因素实验和正交实验确定最优反应条件,并验证其精确度及稳定性。最终确定垃圾降解过程中多酚氧化酶活性测定的最优条件为基质取用10 m L、反应温度为30°C、缓冲液为8 m L、反应时间为1.5 h、萃取剂量为20 m L、萃取时间为50 min,且优化后测定条件实验结果的RSD小于2.22%,测定结果较传统方法提高了41%~90%,说明优化后结果准确可靠,为垃圾中多酚氧化酶的活性测定提供了新的方法基础。 The study optimized the experimental condition of polyphenol oxidase in landfill with the waste sample of bioreactor landfill and on the basis of the soil enzymology. The optimal reaction conditions is determined by single factor experiments and orthogonal experiments,then its accuracy and stability is verified. The results showed that the best experimental conditions of polyphenol oxidase in landfill were as follows: 10 mL of matrix, 30 ℃ of reaction temperature, 8mL of the buffer, 1.5 h of reaction time, 20mL of extraction agen and 50 min of extraction time. The relatively standard deviation (RSD) is below 2.22% when polyphenol oxidase in landfill was measured under these conditions, and the determination results compared with the traditional method has improved by 41% - 90%, the analysis shows that the results are reliable and of high precision, and also provides a new method for the determination of the activity of polyphenol oxidase.
